摘要
目的:研究补中益气汤对膝骨关节炎(KOA)大鼠模型的治疗作用。方法:48只SD大鼠随机分为空白组,模型组,阳性对照组,低、中、高剂量组,每组8只。采用前交叉韧带离断法造模,造模1周后补中益气汤给药,每日2次,持续6周。方法:动物饲养结束后,收集大鼠血清及其整个膝关节。大体观察其关节面的软骨颜色、磨损及缺损程度,HE、阿利新蓝染色观察软骨及软骨细胞改变,Masson染色观察软骨胶原改变,Elisa检测血清中白介素-6(IL-6)、血管内皮生长因子A(VEGFA)的变化,免疫组化检测软骨内IL-6、VEGFA的蛋白表达情况。结果:低、中、高剂量组关节局部损伤均较模型组减轻。Masson染色显示与空白组比较,模型组蓝色胶原显著下调(P<0.01);与模型组比较,阳性对照组及低、中、高剂量组蓝胶原含量均上调(P<0.01,P<0.05)。Elisa结果显示,与空白组比较,模型组IL-6、VEGFA表达均上调(P<0.01);与模型组比较,阳性对照组及中、高剂量组血清IL-6、VEGFA表达均下调(P<0.01),低剂量组血清VEGFA表达显著下调(P<0.05)。免疫组化结果显示,与空白组比较,模型组IL-6、VEGFA表达上调(P<0.01);与模型组比较,阳性对照组、高剂量组IL-6、VEGFA表达均下调(P<0.01,P<0.05)。结论:补中益气汤对KOA大鼠具有治疗作用,其机制可能与该方可降低IL-6、VEGFA的表达有关。
Objective:To study the therapeutic effect of Buzhong Yiqi Decoction on rat model of knee osteoarthritis(KOA).Methods:Forty-eight SD rats were randomly divided into blank group,model group,positive control group,low-dose group,middle-dose group and high-dose group,8 rats in each group.The model was made by amputation of anterior cruciate ligament and Buzhong Yiqi Decoction was given twice a day for six weeks after 1 week of modeling.After the animals were raised,the serum and the whole knee joint of rats were collected.The color,wear and defect degree of articular cartilage were observed.The changes of cartilage and chondrocytes were observed by HE and Alcian blue staining.The changes of cartilage collagen were observed by Masson staining.The changes of IL-6 and VEGFA in serum were detected by Elisa.The expression of IL-6 and VEGFA in cartilage was detected by immunohistochemistry.Results:The local joint injury in the low,middle and high dose groups was less than that in the model group.Masson staining showed that the level of blue collagen in the model group was significantly lower than that in the blank group(P<0.01),to compared with the model group,the contents of blue collagen in positive control group,low dose group,middle dose group and high dose group were all increased(P<0.01,P<0.05).The results of Elisa showed that the expressions of IL-6 and VEGFA in the model group were up-regulated compared with the blank group(P<0.01),to compared with the model group,the expressions of serum IL-6 and VEGFA in the positive group,middle dose group,high dose group were all down-regulated(P<0.01),the expression of VEGFA in the low dose group was down-regulated(P<0.05).The results of immunohistochemistry showed that compared with the blank group,the expression of IL-6 and VEGFA in the model group was up-regulated(P<0.01),to compared with the model group,the expression of IL-6 and VEGFA in the positive control group and high-dose group was slightly down-regulated(P<0.01,P<0.05).Conclusion:Buzhong Yiqi Decoction has therapeutic effect on knee osteoarthritis model rats,and its mechanism may be related to that Buzhong Yiqi Decoction can reduce the expression of IL-6 and VEGFA.
