摘要
[目的]探讨大蒜素对兔膝骨关节炎的作用。[方法]将30只家兔随机分为空白对照组、模型对照组和大蒜素组,每组10只;除空白对照组外其余2组按改良Hulth法建立模型,术后开始灌服药物,大蒜素组(大蒜素,128 mg/kg/d)、空白对照组和模型对照组(等量生理盐水20 ml),连用8周。术后8周处死所有家兔取右膝股骨髁大体观察;苏木精-伊红(HE)和番红O(SA-O)染色对股骨髁关节软骨行Mankin评分;采用q RTPCR法检测股骨内侧髁负重区软骨组织interleukin-1β(白细胞介素-1β,IL-1β)、matrix metalloproteinase-3(基质金属蛋白酶-3,MMP-3)和tissue inhibitor of metalloproteases-1(基质金属蛋白酶组织抑制因子-1,TIMP-1)的m RNA相对表达量。[结果]三组之间比较,空白对照组的Mankin评分最低,IL-1β、MMP-3和TIMP-1 m RNA表达量最低,模型对照组和大蒜素组,分别与空白对照组比较,上述指标差异均有统计学意义(P<0.05);与模型对照组比较,大蒜素组Mankin评分降低,IL-1β、MMP-3、TIMP-1 m RNA表达量降低,差异有统计学意义(P<0.05)。[结论]大蒜素能够改善兔膝骨关节炎Mankin评分并能够下调股骨髁关节软骨中IL-1βm RNA、MMP-3m RNA表达,上调TIMP-1 m RNA表达量。
[Objective] To investigate the effects of allicin on knee osteoarthritis in the rabbit. [Methods] Thirty healthy domestic rabbits were randomly and equally divided into three groups: the blank control group, the model control group and the allicin group, with 10 animals in each group. A rabbit model of knee osteoarthritis was established by the modified Hulth' s method in the model control group and allicin group. After osteoarthritis was induced, allicin was orally adminis- tered in dosage of 128 mg/kg/d in the allicin group, while the same amount of saline (20 ml) was fed in the blank control group and the model control group for 8 weeks. Afterward, the rabbits were sacrificed to harvest the right femoral condyle for gross and histological observation. Safranin O (SA-O) and hematoxyhn-eosin (HE) staining were used to determine the Mankin scores of the articular cartilage. The mRNA expressions of interleukin 1 beta (IL-113), matrix metalloproteinase 3 (MMP-3), and tissue inhibitor of metalloprotease-1 (TIMP-1) were measured by qRT-PCR. [Results ] The blank control group had the lowest Mankin scores, associated with the lowest mRNA expression of IL-113, MMP-3 and TIMP-1, with sta- tistical differences among the 3 groups in all of the aforesaid parameters (P 〈0.05) . Compared with the model control group, the allicin group was proved significantly declined Mankin scores and decreased mRNA expressions of IL- 113, MMP-3 and TIMP-1 (P〈0.05) . [ Conclusions] Allicin improves the Mankin scores in this knee osteoarthritis model of rabbit, associat- ed with down-regulation of the mRNA expressions of IL- 1β and MMP-3, whereas up-regulation the mRNA expression of TIMP-1 in the articular cartilage of femoral condyle.
