RASGRF1和PGAM1在胃癌组织中的表达及与临床病理特征和预后的关系

Expression of RASGRF1 and PGAM1 in gastric cancer tissues and their relationship with clinicopathological characteristics and prognosis of patients

摘要 目的:探讨Ras蛋白特异性鸟苷酸释放因子1(RASGRF1)和磷酸甘油酸变位酶1(PGAM1)在胃癌组织中的表达及其与患者临床病理特征和预后的关系。方法:选取2014年2月-2016年10月于本院行手术治疗的85例胃癌患者为研究对象,实时荧光定量PCR(RT-qPCR)检测胃癌组织和对应的癌旁组织中RASGRF1和PGAM1的mRNA表达,免疫组织化学法检测RASGRF1和PGAM1蛋白表达,分析RASGRF1和PGAM1蛋白表达与患者临床病理特征的关系。Kaplan-Meier法分析RASGRF1和PGAM1蛋白表达与患者预后的关系。Cox比例风险模型分析影响胃癌患者预后的多因素。结果:与癌旁组织比较,胃癌组织中RASGRF1的mRNA和蛋白阳性表达率均显著降低(P<0.05),PGAM1的mRNA和蛋白阳性表达率均显著升高(P<0.05)。胃癌组织中RASGRF1和PGAM1的蛋白表达均与肿瘤分化程度、TNM分期和淋巴结转移相关(P<0.05),而与患者年龄、性别和肿瘤直径无关(P>0.05)。Kaplan-Meier生存分析显示,胃癌组织中RASGRF1阳性表达的患者3年生存率显著高于阴性表达患者(P<0.05),PGAM1阳性表达的患者3年生存率显著低于阴性表达患者(P<0.05)。Cox分析结果显示,RASGRF1和PGAM1均是影响胃癌患者预后的独立因素(P<0.05)。结论:胃癌组织中RASGRF1呈低表达,PGAM1呈高表达,二者均可能参与胃癌的发生发展,有望成为胃癌诊断和预后判断的生物标志物。 Objective:To investigate the expression of Ras protein-specific guanine nucleotide-releasing factor 1(RASGRF1)and phosphoglycerate mutase 1(PGAM1)in gastric cancer tissues and their relationship with the clinicopathological characteristics and prognosis of patients.Methods:A total of 85 gastric cancer patients who underwent surgery in our hospital from February 2014 to October 2016 were selected as the research subjects.Real-time fluorescent quantitative PCR(RT-qPCR)was used to detect the expression of RASGRF1 and PGAM1 mRNA in gastric cancer tissues and corresponding adjacent tissues.Immunohistochemical method was used to detect the expression of RASGRF1 and PGAM1 protein,and the relationship between the expression of RASGRF1 and PGAM1 protein and the clinicopathological characteristics of patients was analyzed.Kaplan-Meier method was used to analyze the relationship between RASGRF1 and PGAM1 protein expression and patient prognosis.The Cox proportional hazards model analyzed multiple factors affecting the prognosis of patients with gastric cancer.Results:Compared with adjacent tissues,the mRNA and protein positive expression rate of RASGRF1 in gastric cancer tissues were significantly reduced(P<0.05),while the mRNA and protein positive expression rate of PGAM1 were significantly increased(P<0.05).The expression of RASGRF1 and PGAM1 protein in gastric cancer tissues were related to tumor differentiation,TNM stage and lymph node metastasis(P<0.05),but were not related to patient age,gender and tumor diameter(P>0.05).Kaplan-Meier analysis showed that the 3-year survival rate of RASGRF1 positive patients was significantly higher than that of negative patients(P<0.05),while the 3-year survival rate of PGAM1 positive patients was significantly lower than that of negative patients(P<0.05).Cox analysis showed that both RASGRF1 and PGAM1 were independent factors affecting the prognosis of patients with gastric cancer(P<0.05).Conclusion:The expression of RASGRF1 is low in gastric cancer tissues,while the expression of PGAM1 is high,both of which are involved in the development of gastric cancer and may become biomarkers for diagnosis and prognosis of gastric cancer.
