摘要
目的探讨厚朴酚对增生性瘢痕成纤维细胞增殖、凋亡和迁移的调节作用。方法自2020年9月至2021年9月,北部战区总医院烧伤整形科将增生性瘢痕成纤维细胞分为4组,分别使用0、10、20和40μmol/L的厚朴酚处理。于作用后0、1、2、3和4 d使用CCK-8试剂盒检测细胞增殖。作用后1 d使用细胞周期试剂盒检测细胞周期情况,细胞凋亡试剂盒检测细胞凋亡水平,台盼蓝染色法检测细胞迁移水平,Western blot实验检测细胞中Cyclin D1、Bcl-2和MMP2的蛋白表达水平。结果细胞增殖实验检测结果显示,与0μmol/L的厚朴酚作用相比,10、20和40μmol/L的厚朴酚作用增生性瘢痕成纤维细胞1~4 d后,能够显著抑制细胞的增殖能力。10、20和40μmol/L的厚朴酚均能够促进G0-G1期细胞的表达,下调S期细胞百分比,抑制细胞周期的进程;促进早期和晚期凋亡细胞的百分比;明显下调增生性瘢痕成纤维细胞的迁移水平。Western blot实验结果显示,厚朴酚可以显著下调细胞中Cyclin D1、Bcl-2和MMP2的蛋白表达水平。结论厚朴酚能够通过抑制增生性瘢痕成纤维细胞的增殖、促进其凋亡、抑制其迁移发挥抑制增生性瘢痕进程的作用。
Objective To explore the effect ofmagnolol on proliferation,apoptosis and migration of hypertrophic scar fibroblasts.Methods From September 2020 to September 2021,the hypertrophic scar fibroblasts were divided into 4 groups.The fibroblasts were treated with magnolol of 0μmol/L,10μmol/L,20μmol/L and 40μmol/L.The proliferation of hypertrophic scar fibroblasts at 0,1,2,3 and4 days after treatment was detected by CCK8 assay.After treated with magnolol of 0μmol/L,10μmol/L,20μmol/L and 40μmol/L for 1 day,the proportion of fibroblasts in different cell cycles was detected by flow cytometry,the apoptosis of fibroblasts was detected by annexin V/propidium iodide,and the migration of fibroblasts was detected by transwell assay.The protein expression level of cyclin D1,Bcl-2 and MMP2 were detected by Western blot.Results Compared with the 0μmol/L group,magnolol of 10μmol/L,20μmol/L and40μmol/L significantly inhibited the proliferation of hypertrophic scar fibroblasts at 1 to 4 days after treatment.Magnolol of 10μmol/L,20μmol/L and 40μmol/L could promote the expression of fibroblasts in G0/G1 phase,down regulate the percentage of fibroblasts in S phase,inhibit the cell cycle progression,promote the percentage of early and late apoptotic cells and down regulate the migration of hypertrophic scar fibroblasts.The results of Western blot showed that magnolol could significantly down regulate the protein expression level of cyclin D1,Bcl-2 and MMP2 in fibroblasts.Conclusion Magnolol can inhibit the hypertrophic scar by inhibiting the proliferation,promoting the apoptosis and inhibiting the migration of hypertrophic scar fibroblasts.
