摘要
目的探讨不同浓度黄芪总黄酮(TFA)对人子宫内膜癌KLE细胞生长的影响。方法培养KLE细胞株,分别加入终浓度为50、100、200、400、800μg/ml的TFA,继续培养12、24、48 h后,通过采用倒置相差显微镜观察、中性红比色法及流式细胞仪监测等方法观察TFA对KLE细胞的形态、生长抑制曲线、细胞周期和凋亡的影响。结果TFA明显抑制KLE细胞增殖,24 h作用最强;流式细胞仪检测显示TFA能使KLE细胞发生G1期阻滞,并诱导KLE细胞凋亡,其作用均随着TFA剂量增加而增强。结论TFA能够抑制KLE细胞增殖,阻滞细胞周期,诱导细胞凋亡。
Objective To investigate the effects of different concentrations of Total Flavonoids of Astragalus(TFA)on the growth of human endometrial cancer KLE cells.Methods KLE cell line was cultured by TFA at different concentrations of 50,100,200,400 and 800μg/ml for 12,24 and 48 h.The effects of TFA on the morphology,growth inhibition curve,cell cycle and apoptosis of KLE cells were observed by inverted phase contrast microscopy,neutral red colorimetry and flow cytometry.Results TFA significantly inhibited the proliferation of KLE cells,with the strongest effect at 24 hours.Flow cytometry revealed that TFA could arrest KLE cells in G1 phase and induce apoptosis of KLE cells,and the effects were enhanced with the increase of TFA concentration.Conclusion TFA can inhibit proliferation,arrest cell cycle and induce apoptosis of KLE ce lls.
