摘要
目的分析子痫前期(PE)患者与正常妊娠孕妇外周血、尿液标本蛋白质组学的差异。方法留取2019年9—12月就诊北京协和医院的PE患者(P组,n=5)和同期正常妊娠的孕妇(C组,n=7)产前、产后24 h外周血清及尿液样本,用质谱技术鉴定样本中的蛋白,采用数据非依赖采集模式(DIA)蛋白质组学定量方法筛选差异蛋白,并对筛选出的差异蛋白进行生物信息学分析。结果通过DIA技术,以组内产前产后对比,筛选出血清中差异蛋白274个,其中P组84个,上调67个,C组190个,上调73个;尿液中差异蛋白671个,其中P组449个,上调224个,C组222个,上调105个。生物信息学分析结果显示,与C组相比较,P组差异蛋白在炎症免疫反应、内皮损伤、细胞增殖和血管发生相关信号通路异常,可以激活肾素-血管紧张素信号通路、肾上腺髓质素信号通路、心肌肥厚信号通路、铁死亡信号通路。结论蛋白质组学研究可以筛选出PE与正常妊娠之间的差异蛋白,这些差异蛋白通过多种信号通路途径参与PE发生、发展及相关损伤。
Objective:To study the proteomics difference of peripheral blood and urine samples between preeclampsia(PE)patients and normal pregnant women.Methods:The serum and urine samples were collected 24 hours before and after delivery from PE patients(group P,n=5)and normal pregnant women(group C,n=7)who were admitted to Peking Union Medical College Hospital from September 2019 to December 2019.The differential proteins were screened by quantitative proteomics method of data independent acquisition(DIA),and the screened differential proteins were analyzed by bioinformatics analysis.Results:Through DIA technology,274 differential proteins were screened in serum by prenatal and postpartum comparison within the group,including 84 in group P with 67 up-regulated,190 in group C with 73 up-regulated.There were 671 differential proteins in urine,including 449 in group P with 224 up-regulated,and 222 in group C with 105 up-regulated.Bioinformatics analysis showed that abnormal differential proteins in group P were related with inflammatory immune response,endothelial injury,cell proliferation and angiogenesis signaling pathway,and which significantly positively activated in the signaling pathway of renin angiotensin,adrenomedullin,myocardial hypertrophy and ferroptosis.Conclusions:The differential proteins between PE and normal pregnancy are screened out by proteomic studies,and these differential proteins are involved in the occurrence&development,and related injury of PE through a variety of signaling pathways.
