摘要
野生菌栽培具有重要的经济价值。本文报道了分离培养野生侧耳属菌株PO1,并进行生物栽培转化的实验结果。以真菌试剂盒提取的PO1基因组DNA为模板,PCR扩增获得了大小为636 bp的ITS序列(GenBank No.MT261808.1)。运用ClustalX和MEGA软件的NJ法构建了侧耳属代表种的系统发育树,结果显示,PO1与NCBI中其他54株侧耳菌株亲缘关系较为密切,相似性达99.04%以上。明确野生侧耳为糙皮侧耳菌株分类后,总结出一套侧耳室内生物栽培转化技术与管理体系,栽培转化的糙皮侧耳PO2头潮菇和二潮菇产量高、口感好,经济效益佳,转化率高达158%,达到实验室侧耳高产栽培的目的,为大棚栽培管理技术提供了重要参考依据。
The cultivation of witd fungi has important economic value.One wild Pleurotus strain was collected for isola-tion,purification and culture,and wild Pleurotus(PO1)was adapted to indoor bag material growth for biological cultivation transformation experiments.The genomic DNA of PO1 was extracted with a fungus kit,and 636 bp ITS sequence was ob-tained by PCR amplification(GenBank accession number MT261808.1).Using the biological analysis software ClustalX and the NJ method in MEGA to construct a phylogenetic tree of the representative species,the results show that PO1 is most close-ly related to other 54 Pleurotus strains,with a similarity of more than 99.04%.After the wild P.ostreatus is classified as P.os-treatus,a set of indoor biological cultivation and transformation technology and management system of P.ostreatus was sum-marized.The P.ostreatus(PO2)head mushroom and the second tide mushrooms transformed by biological cultivation have high yield,good taste and good economic benefits.The conversion rate is as high as 158%,which has achieved the purpose of high-yield cultivation of P.ostreatus in the laboratory,and at the same time provides an important reference for local farmers in greenhouse cultivation and management techniques.
