摘要
目的:采用网络药理学方法探索大青叶治疗呼吸道感染RTI的作用机制及物质基础生物碱。方法:通过TCMSP检索获得的大青叶成分及其作用靶点,通过GeneCards、OMIM、Drugbank数据库预测作用靶点;应用Hiplot平台对交集靶点进行基因本体GO功能富集分析与基因组百科全书KEGG富集分析;运用String数据库和Cytoscape软件构建蛋白相互作用PPI网络和“成分-靶点-通路”网络;采用单因素以及正交试验考察大青叶生物碱提取和含量测定方法。结果:大青叶中的6种生物碱成分,通过作用于PTGS2、ESR1、HSP90AA1、AR、PPARG等靶点,调控IL-17信号通路、钙信号通路、Th17分化细胞、PI3K-Akt等信号通路,发挥抗病毒、抗炎、免疫调节作用,对呼吸道感染产生治疗作用;采用二氯甲烷提取大青叶中总生物碱;酸性染料比色法测定大青叶总生物碱含量,最优条件为:缓冲液pH=3.6、用量为6mL、指示剂用量为4mL、有机溶剂用量为6mL。结论:大青叶中生物碱类成分对RTI相关靶点有潜在作用,能够通过抗病毒、抗炎等改善RTI症状,为后续研究大青叶对RTI的作用机制提供依据。
Objective:To explore the mechanism of Isatis indigotica in treating respiratory tract infection RTI(respiratory tract infection)and its material base alkaloids from molecular network level by using network pharmacology method.Methods:The components and targets of Isatis indigotica were retrieved by TCMSP,and the targets were predicted by GeneCards,OMIM and Drugbank databases.The functional enrichment analysis of gene ontology(GO)and the enrichment analysis of Kyoto encyclopedia of genes and genomes(KEGG)were carried out on the intersection targets by Hiplot platform.Using String database and Cytoscape software to construct PPI network and“component-target-pathway”network of protein interaction;The extraction and content determination methods of alkaloids from Isatidis Folium were investigated by single factor and orthogonal test.Results:Six alkaloids in Isatidis Folium acted on PTGS2,ESR1,HSP90AA1,AR,PPARG and other targets to regulate IL-17signal pathway,calcium signal pathway,Th17differentiated cells,PI3K-Akt and other signal pathways,thus exerting antiviral,anti-inflammatory and immunomodulatory effects,and having therapeutic effects on respiratory tract infection.Extracting total alkaloids from Isatidis Folium with dichloromethane;The acid dye colorimetry was used to determine the total alkaloid content in Isatidis Folium.The optimal conditions were as follows:pH of buffer solution was 3.6,dosage of organic solvent was 6mL,amount of indicator is 4mL,and dosage of organic solvent was 6mL.Conclusion:Alkaloids in Isatidis Folium have potential effects on RTI-related targets,and can improve RTI symptoms through anti-virus and anti-inflammation,which provides a basis for further research on the mechanism of action of Isatidis Folium on RTI.
