太子参参须多糖促进小鼠脾淋巴细胞体外增殖及细胞因子分泌的研究 被引量:12

Study on Radix Pseudostellaria Fibrous Root Polysaccharide Promoting Proliferation and Cytokine Secretion of Mouse Spleen Lymphocytes in Vitro

摘要 设空白对照组及太子参参须多糖(RPFRP)组,给药12.5、25、50、100、200、400μg/mL,分别与小鼠的脾细胞共同培养。经分光光度法筛选RPFRP对小鼠脾淋巴细胞的安全浓度范围,MTT法检测RPFRP对小鼠脾淋巴细胞增殖的影响,流式细胞术检测药物作用后小鼠脾淋巴细胞的凋亡,ELISA法检测药物作用后小鼠脾淋巴细胞上清液中细胞因子含量。结果与空白对照组相比,RPFRP可显著促进小鼠脾淋巴细胞的增殖(P<0.05),当RPFRP质量浓度在12.5μg/mL~400μg/mL时,其协同ConA可显著促进T淋巴细胞的增殖(P<0.05),当RPFRP质量浓度在50μg/mL~400μg/mL时,其协同LPS可显著促进B淋巴细胞的增殖(P<0.05);RPFRP处理组细胞总凋亡率较空白对照组明显下降(P<0.05),当RPFRP质量浓度在25μg/mL~100μg/mL时,处理组细胞的早期凋亡率和晚期凋亡率均显著降低(P<0.05),RPFRP在刺激淋巴细胞分泌细胞因子(IL-2,IL-4,IL-6和IFN-γ)中具有不同程度的促进作用(P>0.05或P<0.05)。RPFRP可促进小鼠脾淋巴细胞增殖,一定质量浓度内能减少脾淋巴细胞的凋亡,促进脾淋巴细胞因子分泌,为RPFRP的开发应用提供参考。 In this study,the blank control group and the RPFRP groups(the drug concentration of 12.5,25,50,100,200,400μg/mL respectively)were set up,and they were co-cultured with mouse spleen cells respectively.A safe concentration range of RPFRP on mouse spleen lymphocytes was screened by spectrophotometry;the effect of RPFRP on the proliferation of mouse spleen lymphocytes was detected by MTT method;the apoptosis of mouse spleen lymphocytes was detected by flow cytometry after drug action;the levels of cytokines in the supernatant of spleen lymphocytes after drug action were detected by ELISA.The results showed that,compared with the blank control group,RPFRP can significantly promote the proliferation of mouse spleen lymphocytes(P<0.05).When the mass concentration of RPFRP is between 12.5-400μg/mL,its synergism with ConA can significantly promote the proliferation of T lymphocytes(P<0.05).When the mass concentration of RPFRP is between 50-400μg/mL,its synergism with LPS can significantly promote the proliferation of B lymphocytes(P<0.05).The total apoptosis rate of RPFRP treatment groups was significantly lower than that of blank control group(P<0.05),when the mass concentration of RPFRP is between 25-100μg/mL,the early and late apoptotic rates in the treatment group were significantly lower than those in the control group(P<0.05).RPFRP has promotion effects in stimulating lymphocytes to secrete cytokines(IL-2,IL-4,IL-6 and IFN-γ)in varying degrees(P>0.05 or P<0.05).The results showed that RPFRP can promote the proliferation of spleen lymphocytes,reduce the apoptosis of spleen lymphocytes in a certain concentration and promote the secretion of cytokines in spleen lymphocytes.This study result can provide reference for the development and application of RPFRP.
