摘要
本研究建立了两条半夏组培苗再生途径:外植体直接诱导再生植株和外植体通过愈伤组织诱导再生植株,之后对两条再生途径产生的组培苗进行综合评价,筛选出较佳的再生途径,并进一步构建半夏组培苗驯化移栽体系,为半夏离体培育及扩繁提供技术支撑。结果表明:通过外植体直接诱导再生植株途径培养的组培苗健壮、次生代谢产物合成能力强、成本较低,更适合农业大规模生产。驯化移栽阶段将半夏组培苗于1/2MS+0.05 mg·L^(-1)IBA培养基中壮苗10 d,之后炼苗4 d移栽于砂质壤土:营养土为1:2的基质中,可显著提高半夏组培苗移栽成活率,且降低了移栽成本,具有良好的实用价值。
In this paper, two regeneration pathways were established, namely, explants directly induce regeneration of plants and explants to induce regeneration of plants through callus. After that, we comprehensively evaluated the tissue culture seedlings produced by the two regeneration pathways to select a better one. Accordingly, the domestication and transplantation system of tissue culture seedlings of Pinellia ternata can be further constructed to provide technical support for in vitro cultivation and propagation of Pinellia ternata. The result shows that the tissue culture seedlings cultivated through the explants directly induce regeneration are more robust and have low costs. Besides, in this way, the seedlings have strong secondary metabolite synthesis ability and are more suitable for large-scale agricultural production. In the domestication and transplanting stage, the Pinellia tissue-cultured seedlings were grown on 1/2 MS+0.05 mg·L^(-1) IBA medium for 10 days, and then the seedlings were refined and transplanted on sandy loam soil for 4 days: the nutrient soil was a 1:2 substrate It can significantly increase the survival rate of transplanting Pinellia tissue culture seedlings and reduce the transplanting cost and has good practical value.
