摘要
以河北杨无菌苗叶片为外植体材料,诱导叶片分化产生不定芽,然后对不定芽进行增殖和生根培养,形成再生植株。结果表明:最佳不定芽诱导培养基为MS+0.3 mg·L^(-1)TDZ+0.5 mg·L^(-1)IBA,不定芽诱导率达到100%;最佳增殖培养基为MS+0.5 mg·L^(-1)KT+0.3 mg·L^(-1)IBA,增殖率可达85%;最佳生根培养基为1/2MS+0.3 mg·L^(-1)IBA,平均生根率可达到92.86%;组培苗移栽成活率为93.7%。本研究建立了河北杨高效植株再生体系,为河北杨的遗传改良和分子育种研究奠定了基础。
The sterile seedlings leaves of Populus hopeiensis were used as explants,the leaves were induced to produce adventitious buds,then the adventitious buds were proliferated and rooted to form regenerated plants.The results showed that the best adventitious bud induction medium was MS+0.3 mg·L^(-1)TDZ+0.5 mg·L^(-1)IBA,the induction rate of adventitious bud was 100%.The best subculture medium was MS+0.5 mg·L^(-1)KT+0.3 mg·L^(-1)IBA,and the proliferation rate was 85%.The best rooting medium was 1/2 MS+0.3 mg·L^(-1)IBA,and the rooting rate was 92.86%.The survival rate of transplanted tissue culture seedlings reached 93.7%.In this study,an efficient regeneration system of Populus hebeiensis was established,laying a foundation for the genetic improvement and molecular breeding research.
