摘要
目的:探究不同环境条件对火炭母提取物的抗氧化性及稳定性的影响。方法:采取体外法测定火炭母提取物对DPPH·、·O_(2)^(-)、·OH三种自由基的清除能力和总还原能力,并分析不同环境条件对其抗氧化性及稳定性的影响。结果:火炭母提取物清除DPPH·的IC_(50)为0.136mg/mL;随着温度的升高,火炭母提取物DPPH·清除能力先升高后降低,变化幅度小,稳定性好;相对于酸性条件,碱性条件下能更好的保持火炭母提取物DPPH·清除能力,但整体清除率降低趋势明显;随着紫外线照射时间的增加,火炭母提取物DPPH·清除能力几乎无变化,耐光性好;蔗糖和柠檬酸钠对火炭母提取物DPPH·清除能力影响较大;金属离子会增强火炭母提取物DPPH·清除能力。结论:火炭母提取物的抗氧化能力较强;温度、紫外线、金属离子对稳定性无影响;酸碱度、蔗糖和柠檬酸钠会影响其稳定性,在实际应用中应避免与此类物质直接接触。
Objective :This study was carried out to investigate the effects of different environmental conditions on stability and antioxidant activity of the extracts from Polygonum chinense L.. Methods :In order to evaluate the antioxidant activity of the extracts from Polygonum chinense L.,the scavenging rates of 1,1-diphenyldiprylhydrazine(DPPH),superoxide anion and hydroxyl radical were determined in vitro. The effects of different environmental conditions on stability and antioxidant activity of the extracts were also investigated. Results:The halfscavenging concentration(IC_(50))was 0.136 mg/m L for scavenging DPPH · by the extract from Polygonum chinense L.. With the increasing of temperature,the scavenge ability of the extracts increased first and then decreased,but the range of changing was small with good stability. By compared with acidic conditions,the extracts had a good stability on scavenging DPPH under alkaline conditions,but the overall removal rate decreased obviously. The stability on scavenging DPPH was stable under ultraviolet radiation by different time. The sucrose and sodium citrate had a great effect on the ability of scavenging DPPH,but the ability could be enhanced by metal ions. Conclusion :The extracts from Polygonum chinense L. had a strong antioxidant capacity. The environmental factors including temperature,ultraviolet rays,and metal ions had no effect on the stability of the extract,but p H,sucrose and sodium citrate might have a great influence on the stability of extracts. Therefore,the direct contacting with these environmental factors should be avoided in practical applications.
