摘要
为了研究头孢噻呋在鸭空肠吸收特征及木犀草素对头孢噻呋在鸭空肠吸收的影响,试验选取9只4周龄的健康鸭只,随机分为3组,分别为对照组(含200μg/mL头孢噻呋)、0.5 MIC木犀草素试验组(含39μg/mL木犀草素的200μg/mL头孢噻呋)和1 MIC木犀草素试验组(含78μg/mL木犀草素的200μg/mL头孢噻呋),利用外翻肠囊法将鸭空肠在各组培养液中进行离体培养。由于头孢噻呋进入肠内会迅速转化为去呋喃甲酰基头孢噻呋(DFC),试验采用高效液相色谱法(HPLC)检测肠囊内液中的DFC,计算头孢噻呋的表观渗透系数(P_(app))、吸收率和吸收速率。结果表明:头孢噻呋在鸭小肠的P_(app)小于1.0×10^(-5) cm/s,表明头孢噻呋在鸭空肠吸收较差;0.5 MIC木犀草素试验组和1 MIC木犀草素试验组中,头孢噻呋的P_(app)、吸收率、吸收速率均显著高于对照组(P<0.05),且1 MIC木犀草素试验组中头孢噻呋的渗透性显著高于0.5 MIC木犀草素试验组(P<0.05)。说明木犀草素能够促进头孢噻呋在鸭空肠的吸收,且随着木犀草素浓度的增加其促进作用增强。
In order to study the absorption characteristics of ceftiofur in duck jejunum and the effect of luteolin on the absorption of ceftiofur in duck jejunum, nine healthy ducks aged four weeks were randomly divided into three groups, control group(containing 200 μg/mL ceftiofur),0.5 MIC luteolin test group(containing 39 μg/mL luteolin in 200 μg/mL ceftiofur) and 1 MIC luteolin test group(containing 78 μg/mL luteolin in 200 μg/mL ceftiofur),respectively. The duck jejunum was cultured in vitro in the medium of each group using the everted gut sac method. Since ceftiofur could be rapidly converted into desfuranyl ceftiofur(DFC) when entering the intestine, high performance liquid chromatography(HPLC) method was used in this experiment to detect the DFC concentration in intestinal cyst fluid;the _(app)arent permeability coefficient(P_(app)), absorption ratio and absorption rate of ceftiofur were calculated. The results showed that the P_(app) of ceftiofur in duck small intestine was less than 1.0×10^(-5) cm/s, which indicated that ceftiofur was poorly absorbed in duck jejunum. In 0.5 MIC luteolin test group and 1 MIC luteolin test group, the P_(app), absorption ratio and absorption rate of ceftiofur were significantly higher than those in control group(P<0.05);the permeability of ceftiofur in 1 MIC luteolin test group was significantly higher than that in 0.5 MIC luteolin test group(P<0.05). The results indicated that luteolin could increase the absorption of ceftiofur in duck intestine, and its promoting effect was enhanced with the rising of luteolin concentration.
