复合植物提取物对湘东黑山羊胃肠道游离氨基酸含量及其转运载体相对表达量的影响 被引量:1

Effects of Plant Extract Mixture on Free Amino Acids Content and Relative Expression Level of Their Transporters in Gastrointestinal Tract of Xiangdong Black Goat

摘要 本试验旨在研究饲粮中添加复合植物(含杜仲叶、迷迭香叶、枇杷叶和柿子叶)提取物对湘东黑山羊胃肠道游离氨基酸含量及其转运载体相对表达量的影响。选择体重相近[(9.82±0.83)kg]的健康雌性湘东黑山羊20头,随机分为2组,每组10头。对照组饲喂基础饲粮,试验组在基础饲粮中添加0.15%的复合植物提取物,试验期56 d。结果表明:与对照组相比,1)饲粮添加复合植物提取物可极显著提高湘东黑山羊瘤胃游离天冬氨酸(Asp)、谷氨酸(Glu)、甘氨酸(Gly)、丙氨酸(Ala)和脯氨酸(Pro)的含量(P<0.01),显著提高游离苏氨酸(Thr)的含量(P<0.05);瘤胃黏膜钙敏感受体(CASR)基因的相对表达量上调了68.6%(P<0.01);2)饲粮添加复合植物提取物可极显著提高湘东黑山羊回肠游离Gly、Ala和精氨酸(Arg)的含量(P<0.01),显著提高游离缬氨酸(Val)的含量(P<0.05);回肠黏膜溶质载体家族1成员1(SLC1A1)基因的相对表达量上调了162.6%(P<0.01),溶质载体家族1成员3(SLC1A3)和溶质载体家族38成员2(SLC38A2)基因的相对表达量分别上调了27.1%和34.3%(P<0.05);3)饲粮添加复合植物提取物可极显著降低盲肠游离Thr、Glu、Gly、酪氨酸(Tyr)和苯丙氨酸(Phe)的含量(P<0.01),同时可显著降低丝氨酸(Ser)、Ala、Val和赖氨酸(Lys)的含量(P<0.05);盲肠黏膜SLC1A1基因的相对表达量下调了77.2%(P<0.01),溶质载体家族7成员1(SLC7A1)基因的相对表达量上调了29.2%(P<0.01);溶质载体家族3成员2(SLC3A2)和SLC38A2基因的相对表达量分别上调了21.5%和34.3%(P<0.05)。综上所述,复合植物提取物能够显著提高湘东黑山羊胃肠道部分游离氨基酸的含量,同时调控胃肠道黏膜氨基酸转运载体的相对表达。 The study was conducted to explore the effects of plant extract mixture(PEM)(Eucommia ulmoides,rosemary,loquat and persimmon leaves)on free amino acids contents and relative expression levels of amino acid transporters in the rumen and intestine of Xiangdong black goats.In total,twenty healthy femalegoats with similar weight of(9.82±0.83)kg were selected and equally randomly assigned into 2 groups,control group and experimental group(basic diets with 0.15%PEM).The experiment lasted for 56 days.The results showed that compared with control group,1)adding PEM to the diet could significantly increase the rumen free aspartic acid(Asp),glutamic acid(Glu),glycine(Gly),alanine(Ala)and proline(Pro)contents of Xiangdong black goats(P<0.01),the content of free threonine(Thr)was significantly increased(P<0.05);the relative expression level of rumen mucosal calcium-sensing receptor(CASR)gene was significantly up-regulated by 68.6%(P<0.01);2)adding PEM to the diet could significantly increase the contents of free Gly,Ala and arginine(Arg)in the ileum of Xiangdong black goats(P<0.01)and significantly increase the content of free valine(Val)(P<0.05);the relative expression level of ileal mucosal solute carrier family 1 member 1(SLC1A1)gene was significantly up-regulated by 162.6%(P<0.01),the relative expression levels of solute carrier family 1 member 3(SLC1A3)and solute carrier family 38 member 2(SLC38A2)genes were significantly increased by 27.1%and 34.3%(P<0.05),respectively;3)adding PEM to the diet could significantly reduce the cecal free Thr,Glu,Gly,tyrosine(Tyr)and phenylalanine(Phe)contents(P<0.01),and at the same time,could significantly reduce serine(Ser)and Ala(P<0.05);the relative expression level of SLC1A1 gene in cecal mucosa was significantly down-regulated by 77.2%(P<0.01),and the relative expression level of solute carrier family 7 member 1(SLC7A1)gene was significantly up-regulated by 29.2%(P<0.01);the relative expression level of solute carrier family 3 member 2(SLC3A2)and SLC38A2 genes were significantly up-regulated by 21.5%and 34.3%(P<0.05).In summary,the PEM can significantly increase the contents of free amino acids in the rumen and intestines of Xiangdong black goats,while regulating the relative expression of amino acid transporters in the rumen and intestinal mucosa.
