摘要
【目的】通过熊果酸(UA)与阿霉素(DOX)的联用,初步探讨其对宫颈癌HeLa细胞增殖的影响,为减轻抗癌药物阿霉素的毒副作用提供依据。【方法】通过细胞试验,采用MTT法检测熊果酸和阿霉素单独及联合使用时的细胞存活率,并筛选出5组不同药物剂量处理:0、20μmol/L UA、2 mg/L DOX、5 mg/L DOX和20μmol/L UA+2 mg/L DOX。根据5组不同处理,采用荧光染色法分析细胞凋亡情况,Western-blot法检测细胞相关凋亡蛋白的表达。【结果】MTT试验结果显示:UA与DOX联用细胞存活率低于各自单独使用的存活率;联用组(20μmol/L UA+2 mg/L DOX)给药处理的HeLa细胞形态变化最为明显,由梭状变为圆形,且细胞凋亡数量高于单独用药组;Western-blot结果显示:Cleaved caspase-3蛋白含量联用组低于5 mg/L DOX组,与20μmoL/L UA组存在显著差异性(P<0.05),但与2 mg/L DOX组不存在显著差异(P>0.05),而Bax/Bcl蛋白含量各处理间不存在显著差异(P>0.05)。【结论】熊果酸与阿霉素联用效果高于2种药物单独使用效果,可以在一定程度上减少阿霉素的使用剂量,增强阿霉素对宫颈癌HeLa细胞的治疗作用。
[Purpose]The combined use of ursolic acid(UA)and doxorubicin(DOX)was used to preliminarily explore its effect on the proliferation of cervical cancer HeLa cells,and provide a basis for reducing the side effects of the anticancer drug doxorubicin.[Methods]Through cell experiments,the MTT method was used to detect the cell survival rate of ursolic acid and doxorubicin when used alone and in combination,and 5 groups of different dosage treatments were screened:0,20μmol/L UA,2 mg/L DOX,5 mg/L DOX and 20μmol/L UA+2 mg/L DOX.According to 5 differ-ent treatments,fluorescence staining was used to analyze cell apoptosis,and Western-blot was used to detect the expression of cell-related apoptotic proteins.[Results]The result of the MTT experiment showed that the cell survival rate of ursolic acid combined with doxorubicin was lower than that of the corresponding drugs alone.The morphology changes of HeLa cells treated with the combination group of 20μmol/L UA+2 mg/L DOX were the most obvious,changing from a spindle shape to a round shape,and the number of apoptosis was higher than that of the single drug group.The Western-blot result showed that the content of caspase-3 protein of combination group was lower than that of 5 mg/L DOX group and different from 20μmol/L UA group(P<0.05),but there was no significant difference with the low-dose doxorubicin group(2 mg/L),and there was no significant difference in Bax/Bcl protein content.[Conclusion]The combination of ursolic acid and doxorubicin can reduce the dosage of doxorubicin to a certain extent and enhance the therapeutic effect of doxorubicin on cer-vical cancer HeLa cells.
