摘要
剑麻溃疡病是近年来在剑麻上发生的一种新病害,为明确剑麻溃疡病的致病菌,本研究通过组织分离纯化和致病性鉴定分离得到致病菌株JM-HN01,运用形态学和分子生物学技术将该菌株鉴定为新暗色柱节孢(Neoscytalidium dimidiatum)。生物学特性研究表明,该菌菌丝生长的最适生长温度为35.0℃,最适pH值为7.0;完全光照条件下有利于菌丝的生长;葡萄糖为碳源最有利于菌丝生长,其次为果糖和蔗糖;牛肉浸膏和胰蛋白胨2种氮源最有利于菌丝生长。本研究给剑麻溃疡病发病规律及防控措施的制定提供一定的理论依据。
The canker disease in sisal is a new pathogen recent years,to clarify the pathogenic bacteria of sisal hemp canker.In this study,one correlative strain JM-HN01 was identified as Neoscytalidium dimidiatum by morphology and molecular biology technology which was isolated and purified from the tissue of sisal.Biological characteristics study showed that the optimum temperature for the mycelial growth was 35.0℃and the optimum pH value for the mycelial growth was 7.0.Full light condition was conducive to the growth of the mycelial.Mycelial growth was the fastest on the medium using glucos,fructose and sucrose as carbon source,or using yeast extract,tryptone as nitrogen source.This study provides a theoretical basis for the pathogenesis of sisal canker and the formulation of control measures.
