杞菊地黄胶囊防治激光性视网膜损伤的作用机制:基于网络药理学探讨

摘要 目的运用网络药理学探讨杞菊地黄胶囊防治激光性视网膜损伤的作用机制。方法基于中药系统药理学数据库,获取杞菊地黄胶囊的活性成分及其潜在靶点,通过口服生物利用度≥30%和类药性≥0.18,筛选出有效成分。在Therapeutic Target Database数据库、Drugbank3.0数据库、DisGeNET 5.0数据库以“laser induced retinal injury”为关键词检索激光性视网膜损伤的潜在靶点,并将潜在靶点导入Uniport数据库进行基因标准化,删除重复及无法标准化靶点基因后整合汇总,作为疾病的潜在靶点基因。运用韦恩图选取药物靶点与疾病靶点的交集作为药效靶点,再将这些药效靶点导入String数据库进行分析,建立蛋白质相互作用核心网络图。对靶点进行京都基因与基因组百科全书、基因本体富集分析。结果通过口服生物利用度和类药性参数筛选后,获得139个活性成分。将活性成分对应的靶点导入Uniprot在线数据库进行基因标准化,得到238个成分靶点。激光性视网膜损伤相关疾病靶点911个,如肿瘤坏死因子、白细胞介素(IL)-6、TP53、IL-10等。基因本体富集分析结果显示,杞菊地黄胶囊治疗激光性视网膜损伤的靶点主要涉及癌症的通路、糖尿病并发症中的AGE-RAGE信号通路、IL-17信号通路、人T细胞白血病病毒1型感染、HIF-1信号通路、铂类耐药性、利什曼病、NF-κB信号通路等。京都基因与基因组百科全书功能注释富集分析结果显示,相关靶点主要富集于糖尿病并发症中的AGE-RAGE信号通路、IL-17信号通路、NF-κB信号通路、HIF-1信号通路等。结论杞菊地黄胶囊通过调节AGE-RAGE信号通路、NF-κB信号通路以及HIF-1信号通路,抑制视网膜内皮细胞的氧化应激反应、炎症反应、免疫反应,从而治疗激光性视网膜损伤。 Objective To explore the mechanism of Qiju Dihuang capsule in preventing and treating laser-induced retinal injury by means of network pharmacology.Methods The active components and potential targets of Qiju Dihuang capsule were obtained based on the systematic pharmacological database of traditional Chinese medicine,and the active components were screened by oral bioavailability≥30%and drug-like≥0.18.The potential targets of laser-induced retinal injury were searched in therapeutic Target Database,Drugbank3.0 database and DisGeNET5.0 database with“laser induced retinal injury”as the keyword,and the potential targets were introduced into Uniport database for gene standardization.After deleting duplicates and unable to standardize target genes,they were integrated and summarized as potential target genes of diseases.Wayne diagram was used to select the intersection of drug targets and disease targets as efficacy targets,and then these efficacy targets were imported into String database for analysis,and the core network map of protein-protein interaction was established.The target was enriched and analyzed by Kyoto Encyclopedia of Gene and Genome and Gene Ontology.Results Totally 139 active components were obtained by screening oral bioavailability and drug-like parameters.The targets corresponding to active components were introduced into Uniprot online database for gene standardization,and 238 component targets were obtained.There were 911 targets related to laser-induced retinal injury,such as tumor necrosis factor,interleukin(IL)-6,TP53,IL-10 and so on.The results of gene ontology enrichment analysis showed that the targets of Qiju Dihuang capsule in the treatment of laser-induced retinal injury were mainly related to the pathway of cancer,AGE-RAGE signal pathway,IL-17 signal pathway,human T cell leukemia virus 1 infection,HIF-1 signal pathway,platinum resistance,leishmaniasis,NF-κB signal pathway and so on.The results of functional annotation enrichment analysis of Kyoto Encyclopedia of Gene and Genome showed that the related targets were mainly concentrated in AGE-RAGE signal pathway,IL-17 signal pathway,NF-κB signal pathway,HIF-1 signal pathway and so on.Conclusion Qiju Dihuang capsule can inhibit oxidative stress,inflammation and immune response of retinal endothelial cells by regulating AGE-RAGE signal pathway,NF-κB signal pathway and HIF-1 signal pathway,thereby treating laser-induced retinal injury.
