摘要
玉米(Zea mays)是世界上最重要的粮食作物之一,研究其抗逆性机理有着十分重要的意义。本研究用在线软件预测了玉米miR408的靶基因,对其中编码锌指蛋白的靶基因Zm ZFP36进行了生物信息学分析,并构建了该基因的植物表达载体。研究结果表明,Zm ZFP36的转录本全长为1169 bp,无内含子,开放阅读框长度为702 bp,编码的蛋白质分子量为24.52 kD;具有亲水性,无跨膜结构域;二级结构包含无规则卷曲、α-螺旋、延伸链和β-转角,所占比例分别为69.10%、20.60%、7.73%和2.58%;系统进化树分析表明ZmZFP36蛋白与小米(Setaria italica)、高粱(Sorghum bicolor)蛋白亲缘关系最近。启动子分析结果表明,它含有低温、干旱及脱落酸等多种非生物胁迫响应元件,有助于后期通过遗传转化的方法进一步研究miR408的调控机理。
Zea mays is one of the most important food crops in the world.It is of great significance to study the resistance mechanism of maize under various stress conditions.In this study,the target genes of miR408 were predicted by online software,and one of the target genes encoding zinc finger protein Zm ZFP36 was cloned.A plant expression vector of Zm ZFP36 gene was constructed.The result showed that the full-length cDNA sequence of Zm ZFP36 gene was 1169 bp.It was a hydrophilic protein containing an open reading frame of 702 bp.The molecular weight was 24.52 kD,without transmembrane domain,and its secondary structure consisted of random coil,alpha-helix and beta turn,which accounted for 69.10%,20.60%,7.73%,and 2.58%,respectively.Phylogenetic analysis showed that the Zm ZFP36 protein was closest to Setaria italica and Sorghum bicolor proteins.Promoter analysis showed that it contained a variety of abiotic stress response elements such as low temperature,drought and ABA stress,which could be helpful for further studying regulation mechanism of miR408 by genetic transformation method.
