吸附树脂分离蛹虫草发酵液中虫草素 被引量:2

Separation of cordycepin from fermentation broth by adsorption resins

摘要 采用吸附法替代离子交换法从蛹虫草发酵液中分离虫草素。首先从5种具有不同孔结构的吸附树脂中筛选出一种吸附容量高(29 mg/g)、容易再生的树脂KA I,随后在pH为5.0时,在25、40和60℃条件下,测定虫草素在KA I上的静态吸附容量,其吸附容量随温度的升高而降低,吸附过程符合Freundlich模型(R 2>0.95);接着采用动态柱吸附脱附的方法分离发酵液中的虫草素,考察流速、高径比(H/D)和温度等工艺条件对虫草素吸附流出曲线的影响;最后采用体积分数50%乙醇对被树脂吸附的虫草素进行洗脱,虫草素收率>97%,洗脱液中虫草素的平均质量浓度为1.22 g/L。洗脱液中的虫草素可通过浓缩、结晶的方法得到,粗品中虫草素的含量约为85%。吸附法分离蛹虫草发酵液中虫草素,简单快捷,适用于大规模生产制备虫草素。 To separate cordycepin from Cordyceps militaris fermentation broth using adsorption method instead of ion exchange,resin KA-I with high adsorption capacity(29 mg/g)and easy regeneration was screened from 5 adsorption resins with different pore structures.The static adsorption capacity of cordycepin on KA-I resin was determined at 25,40,and 60℃and pH 5.0.The isotherm data fitted with Freundlich isotherm equation well(R 2>0.95).Then the dynamic column adsorption and desorption of cordycepin from fermentation broth were carried out and the effect of flow rate,high diameter ratio(H/D)and temperature on the adsorption breakthrough curve of cordycepin was investigated.Finally,50%ethanol was used to elute cordycepin adsorbed on resin KA-I.The recovery of cordycepin was above 97%,and the average concentration of cordycepin in the eluting solution was 1.22 g/L.Cordycepin in the eluent can be obtained by concentration and crystallization.The content of cordycepin in crude products is about 85%.The experimental results show that it is a simple and quick way to isolate cordycepin from fermentation broth by adsorption method,and it is suitable for large-scale production of cordycepin.
