大麻二酚对口腔颌面部炎性痛的镇痛作用及机制研究 被引量:4

摘要 目的:研究大麻二酚(CBD)对口腔颌面部炎性痛的镇痛作用及其可能机制。方法:选取40只C57BL/6J小鼠随机分为对照组、模型组(福尔马林注射诱导炎性痛)、CBD组(30、60 mg/kg)和布洛芬组(n=8),进行口腔颌面部炎性痛造模及给药。以小鼠45 min内累积抓脸时间作为疼痛指标,研究CBD的镇痛作用,并与布洛芬比较。行为学实验后,对唇部组织进行炎性因子的RT-qPCR检测,以及对三叉神经脊束核尾侧亚核(sp5c)的c-Fos表达进行免疫荧光检测。结果:CBD与布洛芬均显著降低二相痛抓脸时间,降低局部炎症因子mRNA水平,下调sp5c的c-Fos阳性神经元表达数目。结论:CBD对口腔颌面部炎性痛具有镇痛效果,其机制可能与下调局部损伤组织炎性因子表达和提高内源性大麻素含量、以及抑制疼痛信号上行传导有关。 Objective:To study the analgesic effect of cannabidiol(CBD)on orofacial inflammatory pain and its mechanism.Methods:40 C57BL/6J mice were randomly divided into 5 groups:control,model(formaline induced inflammatory pain model),CBD(30 mg/kg,60 mg/kg)and ibuprofen(n=8)for modeling inflammatory pain in orofacial area and drug administration.The cumulative face rubbing time within 45 minutes was used as the pain index for the study of the analgesic effect of CBD and comparation with that of ibuprofen.The inflammatory lip tissues were examined by RT-qPCR of the related inflammatory factors and immunofluorescence was adopted to test the c-Fos expression in the trigeminal nucleus caudalis(sp5c).Results:In the model mice,both CBD and ibuprofen significantly reduced the time of face rubbing in phase II,and reduced the mRNA level of inflammatory factors of local tissues,however,only CBD could down-regulate the expression of c-Fos positive neurons in sp5c.Conclusion:CBD has analgesic effect on orofacial inflammatory pain.Its mechanism may involve the down-regulation of inflammatory factors,the up-regulation of endocannabinoids,and the inhibition of the ascending transmission of pain signals.
