瑞士乳杆菌抗高温胁迫补偿应机制 被引量:2

Effects of high temperature stress on the compensation effect mechanism of Lactobaccillus helveticus

摘要 以瑞士乳杆菌NCUF 210.1为实验对象,对其高温胁迫下生长曲线、细胞膜通透性和流动性、可溶性蛋白含量、海藻糖含量和超氧化物歧化酶(SOD)活性多个指标进行测定,并进行转录组测序,对高温胁迫下瑞士乳杆菌的补偿效应进行探究。结果表明,相比最适生长温度(42℃),瑞士乳杆菌在高温胁迫(47℃)下生长受到明显的抑制;细胞膜流动性降低;膜通透性提高;可溶性蛋白含量提高了140.13%(10 h);海藻糖含量提高了17.23%(10 h);SOD活性提高了159.28%(10 h)。转录组测序结果表明,与42℃相比,高温胁迫(47℃)的瑞士乳杆菌可通过促进氨基酸代谢、糖代谢、核苷酸代谢、膜运输和信号传导途径等相关代谢水平,抵抗高温造成的细胞损伤。本研究通过对瑞士乳杆菌进行转录组分析,为研究其在高温胁迫下的分子机制提供参考。 The growth curve,the permeability and fluidity of membrane,the contents of soluble protein and trehaloset,and the superoxide dismutase(SOD)activity of Lactobaccillus helveticus under high temperature stress were studied.The transcriptome sequencing was also conducted to explore the compensation effect of L.helveticus under high temperature stress.The results showed that,compared with the optimal temperature(42℃),the growth of L.helveticus was significantly inhibited under high temperature stress(47℃),the membrane fluidity of L.helveticus was decreased and the membrane permeability of L.helveticus was elevated,the soluble protein content of L.helveticus increased by 140.13%(10 h),the trehalose content of L.helveticus increased by 17.23%(10 h),and the SOD activity of L.helveticus increased by 159.28%(10 h).Transcriptome sequencing results showed that the cell of L.helveticus under high temperature stress may resist the heat injury by promoting the metaboilic level of amino acid,carbohydrate,nucleotide,membrane transport and signal transduction.This study provides the insights into the heat tolerance mechanism of L.helveticus.
