响应面优化紫油厚朴挥发油提取及抑菌性研究 被引量:5

Optimization of volatil oil extraction from Magnolia officinalis Rehd.et Wils.by response surface methodology and its antibacterial activity

摘要 以湖北恩施特有的紫油厚朴为原料,考察浸泡时间、液料比及提取时间对挥发油得率的影响。利用单因素试验与响应面分析,确定最优工艺参数,并采用牛津杯法研究厚朴挥发油对3种常见致病菌的抑菌活性。结果表明:厚朴挥发油的最佳提取工艺参数为浸泡时间100 min、液料比8.5∶1(mL/g)、提取时间4.5 h,在此条件下,厚朴挥发油提取率为0.187%±0.002%。厚朴挥发油对所选3种致病菌均有较好的抑制作用,尤其是金黄色葡萄球菌,抑菌圈直径达到(45.54±0.47)mm。提取方法操作简单、结果稳定,且提取物具有一定的抑菌性能,为厚朴的进一步开发提供科学依据。 The effects of the type of soaking time,liquid-solid ratio and extraction time on the degree of volatile oil yield were investigated using Magnolia officinalis Rehd.et Wils.from Enshi as raw material.Single factor test and response surface analysis were used to determine the optimal process parameters,and Oxford-cup method was used to study the antibacterial activity of Magnolia officinalis Rehd.et Wils.volatile oil against three common pathogens.The results showed that the optimal process parameters of extraction for volatile oil from Magnolia officinalis Rehd.et Wils.was founded to be soaking time of 100 min,liquid-solid ratio of 8.5∶1(mL/g)and extraction time of 4.5 h.Under these conditions,the actual volatile oil yield was 0.187%±0.002%.The volatile oil from Magnolia officinalis Rehd.et Wils.had certain inhibitory effect on three common pathogenic bacteria,especial in inhibiting Staphylococcus aureus,the bacteriostatic circle diameter were(45.54±0.47)mm.The extraction had a strong broadspectrum antibacterial effect which provide a scientific basis in order to deeply research volatile oil from Magnolia officinalis Rehd.et Wils.
