摘要
目的建立柱前衍生-高效液相色谱法测定酒中氰化物含量的方法。方法采用异烟酸-巴比妥酸柱前衍生,高效液相色谱法测定。采用菲罗门Gemini C18(4.6 mm×250 mm,5μm)色谱柱,以1%乙酸-乙腈(50:50)为流动相,流速为1.0 ml/min,检测波长为600 nm。结果氰化物的衍生产物的线性范围为0.5~40μg/L,相关系数大于0.9998。仪器检出限为0.5μg/L,白酒、啤酒、葡萄酒中氰化物的平均回收率分别为96.7%,95.7%,96.3%(n=9)。结论该方法准确可靠、灵敏度高、稳定性好,有较好的样品适用性,可为酒中氰化物的测定标准提升提供参考。
Objective To establish a pre-column derivatization HPLC method for the determination of cyanide in liquor.Methods An HPLC method was applied,using isonicotinic acid-barbituric acid as precolumn derivatization agent.Phenomenex Gemini C18(4.6 mm×250 mm,5μm)column was adopted,using 1%acetic acid-acetonitrile(50:50)at the flow rate of 1.0 ml/min as mobile phase.The detection wavelength was set at 600 nm.Results The calibration curve was linear over the concentration range of 0.5-40μg/L for cyanide derivatives.The correlation coefficients of cyanide derivatives were greater than 0.9998.The limit of detection was 0.5μg/L.The average recoveries of cyanide were 96.7%for white spirits,95.7%for beer and 96.3%for wine(n=9).Conclusion The developed HPLC method is accurate,sensitive,stable and has good sample applicability.It can supply reference for improvement of the determination standard of cyanide in liquor.
