摘要
目的探讨miR-429对甲状腺癌细胞增殖迁移能力的影响及其作用机制.方法将甲状腺癌细胞分为观察组(anti-miR-429质粒转染)、阴性对照组(miR-429空载体质粒转染)、空白对照组(不进行转染),分别进行CCK-8细胞增殖实验、Transwell细胞迁移和侵袭实验.RT-PCR法检测miR-429表达水平,Western blot法检测MMP-2、MMP-9、P21、CyclinD1蛋白表达水平.结果转染48 h后,观察组SW579细胞中miR-429表达水平明显低于阴性对照组和空白对照组,差异具有统计学意义(P<0.01).SW579细胞培养12、24 h时,增殖水平在三组间比较差异无统计学意义(P>0.05);SW579细胞培养48、72 h时,观察组明显高于阴性对照组和空白对照组,差异具有统计学意义(P<0.05).观察组迁移细胞数和侵袭细胞数均明显多于阴性对照组和空白对照组,差异具有统计学意义(P<0.01).观察组MMP-2、MMP-9、P21、CyclinD1蛋白相对表达量明显高于阴性对照组和空白对照组,差异具有统计学意义(P<0.01).结论miR-429在甲状腺癌中是一种抑癌基因,能够有效抑制甲状腺癌细胞的增殖和分化,可能是通过MMP-2、MMP-9、P21、CyclinD1蛋白而发挥抑癌基因的作用.
Objective To investigate the effect of miR-429 on the proliferation,migration and invasion of thyroid cancer cells.Method Thyroid cancer cells were divided into three groups:observation group(anti-miR-429 plasmid transfection),negative control group(miR-429 plasmid transfection),blank control group(no transfection).CCK-8 cell proliferation test,Transwell cell migration and invasion test were performed.RT-PCR was used to detect miR-429 expression level,and Western blot was used to detect MMP-2,MMP-9,P21,CyclinD1 protein levels.Results 48 hours after transfection,the expression level of miR-429 in SW579 cells in the observation group was significantly lower than that in the negative control group and blank control group(P<0.01).There was no significant difference in the proliferation level of SW579 cells among the three groups at 12 h and 24 h(P>0.05);The proliferation level of SW579 cells in the observation group was significantly higher than that in the negative control group and blank control group at 48 h and 72 h(P<0.05).The number of migration cells and invasive cells in the observation group were significantly more than those in the negative control group and the blank control group(P<0.01).The relative expression of MMP-2,MMP-9,P21,CyclinD1 protein in the observation group was significantly higher than that in the negative control group and blank control group(P<0.01).Conclusion miR-429 is a tumor suppressor gene in thyroid cancer,which can effectively inhibit the proliferation and differentiation of thyroid cancer cells.miR-429 may play the role of tumor suppressor gene through MMP-2,MMP-9,P21,CyclinD1 proteins.
