摘要
以梭子蟹蟹壳为原料,经预处理、浸酸脱钙质、碱煮脱蛋白和浓碱脱乙酰化制备壳聚糖膜;再在碱性条件下使壳聚糖膜和氯乙酸发生取代反应,制备羧甲基化壳聚糖膜。采用电位滴定法测试羧甲基化壳聚糖膜的取代度。测试壳聚糖膜和羧甲基化壳聚糖膜的溶胀性、吸水性、润湿性和抗菌性。结果表明:制备的羧甲基化壳聚糖膜的取代度为1.86,其溶胀性和吸水性均优于壳聚糖膜,表面润湿性好,对大肠埃希菌有一定的抑制效果。该研究结果可为壳聚糖基医用补片生物材料的进一步开发提供理论基础。
Using portunus crab shell as raw material,chitosan membranes were prepared by pretreatment,acid soaking decalcification,dilute alkali deproteinization and concentrated alkali deacetylation.Then carboxymethylated chitosan membranes were prepared by substitution reaction between chitosan membranes and chloroacetic acid under alkaline conditions.The degree of substitution of carboxymethylated chitosan membranes was measured by potentiometric titration.The swelling,water absorption,wettability and antibacterial properties of the chitosan membranes and the carboxymethylated chitosan membranes were tested.The results showed that,the prepared carboxymethylated chitosan membrane had a degree of substitution of 1.86,swelling and water absorption properties were better than those of chitosan membranes,surface wettability was superior,and had a certain inhibitory effect on Escherichia coli.The results could provide a theoretical basis for the further development of chitosan-based biomaterials for medical patches.
