Growth and Adaptability of Transgenic Populus simonii×P. nigra with betA Gene

摘要 为了选出速生耐盐的转betA基因小黑杨优良株系,以盐碱地营建的13年生转betA基因小黑杨试验林为材料,开展生长适应性分析,结合5年生时该试验林优良株系选择结果,对早期评价进行验证。结果显示:参试株系树高、胸径、材积生长及保存率等性状在株系间的差异均达到极显著水平(P<0.01),综合评价选出T01、T06和T08株系为优良株系,其中T01株系的树高、胸径和材积均值分别为11.03 m、11.76 cm和0.055 m3,较对照株系分别高16.42%、14.81%和48.67%。该试验林本次选择结果与5年生时的选择结果一致,说明转betA小黑杨在造林试验初期进行优良株系选择也准确可靠。参试株系分子检测显示,外源betA基因仍稳定整合于转betA小黑杨的基因组中,并且能够正常表达。入选的T01、T06和T08株系为生长量大、耐盐性强的优良株系,这些入选株系为后续转betA小黑杨的环境释放及生产性试验提供指导。 In order to select the excellent lines of fast-growing and salt-tolerant transgenic Populus simonii×P. nigra, the 13-year-old BetA gene Populus simonii×P. nigra experimental forest constructed in saline-alkali soil was used as the material to carry out growth adaptation analysis, combined with the excellent lines of the experimental forest at 5 years. The results were selected to verify the early evaluation and molecular detection was performed on the three selected elite transgenic lines. The differences in tree height, DBH, volume and preservation rate of the tested lines reached a very significant level(P<0.01). Through comprehensive evaluation, T01, T06 and T08 lines were selected as elite lines. The average tree height, DBH and volume of the optimal line(T01) were 11.03 m, 11.76 cm and 0.055 m3, respectively. They were higher than 16.42%, 14.81% and 48.67% of the control line(CK), respectively. The selected lines were consistent with the selection of the elite lines of the experimental forest at the time of 5-year-old, indicating that the selection of excellent lines in the early stage of the afforestation trial was also accurate and reliable. The molecular detection results of the remaining lines showed that the exogenous betA gene in the trans-betA. sinensis was stably integrated into the genome chromosome and could be expressed normally. The selected T01, T06 and T08 lines are elite lines with large growth and salt tolerance. These selected strains would provide guidance for the environmental release and production test of the subsequent transfer of betA.
