广西猪源H9N2亚型流感病毒的分离与鉴定及HA、NA基因序列分析 被引量:4

Isolation and identification of swine-origined influenza virus H9N2 subtype isolates from Guangxi Province

摘要 为了解广西猪源H9N2亚型流感病毒的流行情况,试验用某猪场送检的疑似流感病料接种9~10日龄SPF鸡胚尿囊腔进行病毒分离,采用RT-PCR及血清学方法对分离毒株进行鉴定,PCR扩增分离毒株的部分HA、NA基因,并对其进行测序分析。结果表明:从疑似流感病料中成功分离出2株猪源流感病毒,将其命名为A/swine/Guangxi/P2/2011和A/swine/Guangxi/P3/2011;2株分离毒株血凝(HA)试验的血凝价为1∶27,与H9亚型流感病毒阳性血清的血凝抑制(HI)试验的血凝价为1∶27,而与其他亚型流感病毒及新城疫病毒(NDV)阳性血清均没有发生抑制反应;在交叉神经氨酸酶抑制试验(NI)中,2株分离毒株只被N2亚型阳性血清抑制,而与其余7种亚型阳性血清均没有发生抑制反应;部分HA基因与H9亚型流感病毒核苷酸序列同源性最高,为81.1%~92.2%;部分NA基因与N2亚型流感病毒核苷酸序列同源性最高,为77.3%~88.7%。说明试验分离得到的2株猪源流感病毒株均属于H9N2亚型。 In order to understand the prevalence of H9 N2 subtype influenza virus from pigs in Guangxi Province, the suspected influenza virus samples sent by a pig farm were inoculated the allantoic cavity of SPF chick embryos aged 9-10 days to isolate the virus. RT-PCR and serology methods were used for the identification of the isolated strains. Partial HA and NA genes of the isolated strains were amplified and sequenced. The results showed that 2 strains of swine-origined influenza virus were successfully isolated from the suspected influenza material samples, and were named A/swine/Guangxi/P2/2011 and A/swine/Guangxi/P3/2011. The hemagglutination(HA) values of the two isolated strains were 1∶27,and the hemagglutination inhibition(HI) test of the two isolated strains reacted with the H9 subtype influenza virus positive serum were 1∶27,while those isolates had no inhibitory reaction with the positive serum of other Subtypes influenza virus and Newcastle disease virus(NDV) in the HI test. In the cross-neuraminidase inhibition test(NI),the two isolated strains were only inhibited by the N2 subtype positive sera, and had no inhibitory reaction with the other 8 subtypes positive sera. Some HA genes had the highest nucleotide sequence homology with H9 subtype influenza virus, ranging from 81.1% to 92.2%;some NA genes had the highest nucleotide sequence homology with N2 subtype influenza virus, ranging from 77.3% to 88.7%. The results suggested that the two swine-origined influenza virus isolates belonged to the H9 N2 subtype.
