摘要
狭叶泥炭藓属于泥炭藓科,一般生长在高山沼泽湿地环境中,属于这类环境中的特有群落,在此类生境生物演替中是不可替代的,其生态作用具有持水性强的特点。目前泥炭藓湿地群落遭到严重破坏,狭叶泥炭藓生长缓慢且生长周期长,通过组织培养技术可以扩大生产。为了能够快速培养出狭叶泥炭藓的无菌苗,本研究选取贵州省独山县甲定乡的狭叶泥炭藓作为试验材料,发现狭叶泥炭藓的最适消毒方法:用洗衣粉水浸泡5~10 min预处理,流水冲洗2 h左右;转入超净工作台用75%酒精浸泡狭叶泥炭藓外植体60 s,无菌水冲洗7~8次;接着用0.35%的次氯酸钠溶液浸泡55 s,无菌水冲洗6~7次,此组合未污染率最高达70%,成活率最高为82%。通过组织培养,筛选最适合狭叶泥炭藓生长的培养基,在Knop、Beneck、BG-11以及MS 4种培养基中Beneck培养基的发芽率最好、新生配子体的株高最高,最适合狭叶泥炭藓的生长;在Bneneck基本培养基下探究了pH值和IAA对狭叶泥炭藓生长的影响,发现Bneneck培养基pH值为5~6时最适狭叶泥炭藓生长,培养基添加10-4 mol/L的IAA对无菌苗的生长高度和新增分枝长度有明显的促进作用。本研究可为狭叶泥炭藓的进一步研究提供基础数据和资料。
Sphagnun cuspidatum belongs to the Sphagnaceae and generally grows in alpine marsh wetland environments.It is a unique community in this type of environment.It is irreplaceable in this type of habitat biological succession,and it has ecological effect of strong water holding capacity.The peat%Sphagnun wetland community is currently severely damaged.Sphagnum cuspidatum grows slowly and has a long growth period.Tissue culture technology can be used to expand production.This research selected Sphagnum cuspidatum as the experimental material in Jiading Township,Dushan County,Guizhou Province,and discovered the most suitable disinfection method for Sphagnum cuspidatum:Soaked in washing powder water for 5~10 min for pretreatment,and washed under running water for about 2 h.Transfer to a clean bench and soak the Sphagnum cuspidatum explants with 75%alcohol for 60 s,and rinse 7~8 times with sterile water.Then,immerse it in 0.35%sodium hypochlorite solution for 55 s,and rinse 6~7 times with sterile water,and the combined uncontamination rate is up to 70%,and the survival rate is up to 82%.Tissue culture was used to select the best medium for the growth of Sphagnum cuspida-%tum.Among the four types of media,Knop,Beneck,BG-11 and MS,Beneck's medium had the best germination rate and the highest plant height of the new gametophyte.The effects of pH and IAA on the growth of Sphagnum cuspidatum were explored in Bneneck basic medium.Optimal growth of Sphagnum angustifolia when the pH of the Bneneck medium was 5~6.Adding 10-4 mol/L IAA can significantly promote the growth height and the length of new branches of aseptic seedlings.This research can provide basic data and information for further research of Sphagnum cuspidatum.
