摘要
小立碗藓(Physcomitrella patens(Hewd.)B.S.G.)作为独特的模式植物在分子生物学研究中被广泛应用.本实验以改良的Knop培养基为基本培养基,研究添加不同种类和浓度配比的植物激素对小立碗藓愈伤组织诱导及分化的影响.实验结果表明:当糖浓度为2%,添加KT 0.05 mg·L^(-1)或添加6-BA 0.05 mg·L^(-1),在温度(25±1)℃,光照强度3000~4000lx,光照周期12/12 h/d条件下培养,诱导小立碗藓愈伤组织的效果最为理想.诱导愈伤组织分化的实验结果表明:未添加任何植物激素的Knop培养基上分化出正常的配子体且数量最多.添加2,4-D或IAA的培养基上能长出较多的原丝体.
The moss Physcomitrella patens (Hedw) B. S.G. as a model and unique experimental material has been widely used in molecular studies. In present research, we cultured Physcomitrella patens's tine of stems on the improved medium Knop plus different kinds and concentrations of plant hormones, at temperature ( 25 ± 1 ) ℃, light intensity 3000 -40001x, and 12h day/ 12h night. Calli of Physcomitrella patens were induced better in medium Knop plus 6-BA 0.05 mg·L^-1 or KT 0.05mg · L^-1 when the concentration of glucose is 2%. Then took calli of Physcomitrella patens on the media plus different plant hormones, normal gametophyte of Physcomitrella patens were differentiated in medium Knop. Moreover, protonemata were gained in medium Knop added 2,4-D or IAA.
