摘要
【目的】确定草鱼(Ctenopharyngodon idellus)中出现的以肌肉出血、脾脏肿大、肠炎、鳔出血为典型症状的疾病的病原,为该病的防治提供参考。【方法】采用病原分离纯化、生理生化试验、16S r DNA检测、系统进化分析、动物回归感染试验等方法对致病菌进行鉴定,采用纸片法确定病原的药敏特性。【结果】分离株为革兰氏阴性短杆菌,菌落表面光滑、圆形,颜色为白色;有运动性,硫化氢反应、V-P反应阴性,能发酵葡萄糖、果糖、麦芽糖;16S r DNA测序结果与鲁氏耶尔森菌(Yersinia ruckeri)同源性为99%;回归试验攻毒组与临床病例表现一致;药敏试验结果显示该菌对氨苄西林、恩诺沙星、氟苯尼考等19种抗菌药敏感,对青霉素G、苯唑西林、利福平、万古霉素、克拉霉素、红霉素、头孢他啶、麦迪霉素和痢特灵存在耐药性。【结论】该例草鱼疾病的病原为鲁氏耶尔森菌,在草鱼上为首次报道,生产中可选用恩诺沙星、氟苯尼考等水产类常用抗菌药进行防治。
【Objective】The research was conducted to clarify the etiology of grass carp(Ctenopharyngodon idellus)manifested as muscle hem-orrhage,splenomegaly enteritis and swim bladder hemorrhage,provide reference for the disease prevention and treatment.【Method】Bacterial separation and purification,physiological and biochemical tests,16S rDNA detection,phylogenetic analysis and animal regression test were carried out to identify the pathogen.The drug sensitivity test was carried out by disk diffusion method.【Result】The results showed that the isolate was gram-negative brevibacterium,the surface of the colony was smooth and round,and the color was white.The bacteria were motili-ty,the hydrogen sulfide and V-P reaction were negative,and the glucose,fructose and maltose could be fermented.The homology of 16S rD-NA amplificated by PCR was 99%to Yersinia ruckeri.The artificial infection symptoms was consistent with the clinical case.The results of drug susceptibility test showed that the bacteria were sensitive to 19 kinds of antibacterial drugs such as ampicillin,enrofloxacin and florfeni-col,but not sensitive to penicillin G,oxacillin,rifampicin,vancomycin,clarithromycin,erythromycin ceftazidime,medimycin and furazoli-din.【Conclusion】The pathogen of this case of grass carp disease was Yersinia ruckeri,which was reported for the first time in grass carp.The common antibacterial drugs such as enrofloxacin and florfenicol can be used to prevent and treat the disease.
