摘要
目的探究芒果苷对脂多糖诱导小鼠成骨细胞凋亡的影响及可能机制。方法将小鼠成骨细胞株MC3T3-E1细胞分为对照组、LPS组和LPS+芒果苷组,分别加入PBS缓冲液、脂多糖(LPS)、LPS+芒果苷。采用MTT法检测不同处理因素下各组成骨细胞的细胞存活率;qRTPCR法检测各组细胞炎症因子TNF-α及细胞凋亡相关因子Bax、Bcl-2和Caspase-3基因mRNA的表达。结果与对照组相比,LPS组MC3T3-E1细胞存活率明显降低,TNF-α、Bax和Caspase-3的mRNA表达明显升高,Bcl-2 mRNA表达明显下降;而与LPS组相比,LPS+芒果苷组MC3T3-E1细胞存活率明显提高,TNF-α、Bax、Caspase-3 mRNA表达明显下降,Bcl-2 mRNA基因表达明显升高,各组间差异均有统计学意义(P<0.05)。结论芒果苷可改善LPS导致的成骨细胞凋亡,其作用机制可能与芒果苷抑制LPS诱导的TNF-α形成及调节细胞内Bax、Bcl-2、Caspase-3产生有关。
Objective To explore the effect of mangiferin on apoptosis of MC3T3-E1 osteoblasts induced by lipopolysaccharide and the potential mechanism.Methods The mouse osteogenic cell line MC3T3-E1 was divided into control group,LPS group and LPS+mangiferin group,and cocultured with PBS buffer,lipopolysaccharide(LPS),or LPS+mangiferin,respectively.Then,the survival of cells was detected using MTT method.The mRNA expression of TNF-α,Bax,Bcl-2 and Caspase-3 was detected by RT-PCR.Results Compared with the control group,the survival rate of MC3T3-E1 cells in LPS group was decreased,and the mRNA expression of TNF-α,Bax and Caspase-3 was increased,and the mRNA expression of Bcl-2 was down-regulated.Compared with the LPS group,the MC3T3-E1 cell survival rate of LPS+mangiferin group was improved,the mRNA expression of TNF-α,Bax,Caspase-3 was down-regulated,and the mRNA expression of Bcl-2 expression was up-regulated,the results showed statistically significant differences among different groups(P<0.05).Conclusion Mangiferin can reverse the LPS induced apoptosis of osteoblasts.The potential mechanism may be that mangiferin can inhibit the expression of TNF-αand regulate the production of Bax,Bcl-2,and Caspase-3 in cells.
