摘要
目的:探究天麻粉对慢性不可预见应激(CUMS)抑郁模型小鼠的抗抑郁作用以及对小鼠海马区病理形态的影响。方法:将48只雄性小鼠随机分为对照组,模型组,阳性组,天麻粉低、中、高剂量组,每组8只。除对照组外,其余各组采用慢性不可预见应激法建立抑郁症模型,造模时间为21天。对照组和模型组灌服生理盐水(0.01 ml/g),阳性组灌服阿米替林(15 mg/kg),天麻粉低、中、高剂量灌服不同剂量的天麻粉(0.4g/kg、0.8g/kg)、第43天,第44天通过旷场实验以及悬尾实验检测各组小鼠的抑郁样行为,行为学检测结束后麻醉小鼠,取小鼠大脑组织,通过HE染色和尼氏染色观察海马CA1区的病理形态。结果:(1)慢性不可预见应激(CUMS)可以导致明显的抑郁样行为,包括糖水偏好减少(P<0.01),旷场实验总路程增加(P<0.05)以及悬尾实验不动时间减少(P<0.01)。(2)与模型组相比,天麻粉低、中、高剂量给药组能明显改善抑郁小鼠的糖水偏爱程度,以及矿场实验总路程增加。天麻粉高剂量组能明显缩短悬尾实验不动时间。(3)与空白组相比,模型组小鼠海马CA1区细胞明显凋亡,排列不规则,染色颜色明显加深。与模型组相比,天麻粉各给药组凋亡细胞明显减少,可见大量正常细胞。结论:天麻粉对CUMS模型小鼠具有抗抑郁作用,且对海马区的神经细胞具有一定的保护作用。
Objective: To investigate the antidepressant effect of Gastrodia elata powder on chronic unpredictable stress(CUMS) depression model mice and the effect on the pathological morphology of hippocampus of mice. Method: 48 male mice were randomly divided into control group, model group, positive group and Gastrodia elata powder low, medium and high dose group with 8 rats in each group. Except the control group, the other groups used chronic unpredictable stress method to establish the depression model, the modeling time was 21 d. Normal saline(0.01 ml/g), amitriptyline(15 mg/kg) and low, medium and high doses of Gastrodia elata powder(0.4 g/kg, 0.8 g/kg, 1.2 g/kg) were administered in the control group and model group for 21 d. On the day before the experiment, 21 st day, 42 nd day, the sugar water preference of each group was detected, and on the 43 rd day, 44 th day, the depression-like behavior of each group was detected by open field experiment and tail suspension experiment. The pathological morphology of hippocampal CA1 area was observed by HE staining and Nissler staining. Results:(1) Chronic unpredictable stress(CUMS) can lead to significant depression-like behaviors, including decreased sugar-water preference(P<0.01), increased total distance in open-field experiments(P<0.05) and decreased immobility time in tail suspension experiments(P<0.01).(2) Compared with the model group, the low, medium and high doses of Gastrodia elata powder could obviously improve the sugar and water preference of depressed mice, and the total distance of field experiment was increased. Gastrodia elata powder high dose group can obviously shorten the time of suspension tail experiment.(3) Compared with the blank group, the cells in the hippocampal CA1 area of the model group were obviously apoptotic, irregular in arrangement, and the staining color was obviously deepened. Compared with the model group, the apoptotic cells in each administration group of Gastrodia elata powder decreased significantly, and a large number of normal cells were seen. Conclusion: Gastrodia elata powder has antidepressant effect on CUMS model mice and protective effect on hippocampal neurons.
