摘要
海藻糖是昆虫血淋巴中最重要的糖类物质,海藻糖合成酶(trehalose-6-phosphate synthase,TPS)是昆虫体内海藻糖合成途径的关键酶。本试验通过转录组测序获得一条大豆蚜Aphis glycines(Matsumura)海藻糖合成酶的cDNA序列,命名为AgTPS(GenBank登录号:MN068811)。cDNA序列全长4174 bp,包含一个长度为2559 bp的开放阅读框,编码852个氨基酸,等电点为5.98,分子质量为95.8 kDa,具有一个典型的TPS结构域。不同温度下AgTPS基因干扰结果显示:5℃时基因沉默持续时间最长,30℃处理6 h时该基因沉默效果最好,抑制率可达79.48%,且30℃干扰后对大豆蚜生存和繁殖影响最大。试验表明该基因参与大豆蚜抗逆过程,试验结果为利用RNA干扰技术防治大豆蚜提供理论依据。
Trehalose is the most important carbohydrate in insect hemolymph,and trehalose-6-phosphate synthase(TPS)is a key enzyme in the trehalose synthesis pathway in insects.In this study,a cDNA sequence of soybean aphid Aphis glycines(Matsumura)trehalose synthase was obtained through transcriptome sequencing,and named AgTPS(GenBank accession number:MN068811).The cDNA sequence is 4174 bp in length,contains an open reading frame of 2559 bp and encodes 852 amino acids.The isoelectric point of the deduced amino acid is 5.98.The molecular mass is 95.8 kDa.The deduced amino acid has a typical TPS domain.AgTPS was inhibited at different temperatures.It was at 5℃that AgTPS silenced for a long duration.When inhibited at 30℃for 6 h,the gene had the best silence effect and the inhibition rate was 79.48%.The survival and reproduction of soybean aphid were also most affected at 30℃.The results indicated that AgTPS was involved in the stress resistance of soybean aphid.This study provided theoretical basis for the prevention and control of soybean aphid by RNA interference technology.
