摘要
目的建立紫连油的HPLC指纹图谱和含量测定方法,为紫连油的质量控制与评价提供依据。方法采用薄层色谱法对紫连油中紫草、黄连、冰片进行定性鉴别;采用HPLC法测定不同批次紫连油的指纹图谱,并测定β,β’-二甲基丙烯酰阿卡宁含量。结果薄层色谱法鉴别显示,紫草、黄连、冰片斑点清晰,专属性好;通过HPLC指纹图谱分析,标定了紫连油的13个共有峰,并确认了盐酸小檗碱和β,β’-二甲基丙烯酰阿卡宁的特征峰,各批次间的指纹图谱相似度≥0.981;β,β’-二甲基丙烯酰阿卡宁在40.48~202.40 μg范围内线性关系良好。结论本研究建立的薄层色谱鉴别、指纹图谱和含量测定方法简单、可靠,稳定性好,可为紫连油的质量控制提供参考。
Objective To establish HPLC fingerprint and content determination methods of Zilian oil,which will provide the scientific basis for the quality control and evaluation of Zilian oil.Methods TLC was used for the qualitative identification of Arnebiae radix,Coptidis rhizoma and Borneolum syntheticum in oil.HPLC was adopted for to determine the fingerprints of different batches,and the content determination ofβ,β’-Dimethylacrylalkannin of Zilian oil was determined.Results TLC showed that Arnebiae radix,Coptidis rhizoma and Borneolum syntheticum had clear spots and good specificity.A total of 13 common peaks of Zilian oil were identified by HPLC fingerprint analysis,and 2 characteristic peaks(Berberine hydrochloride andβ,β’-Dimethylacrylalkannin)were identified,and the similarity of fingerprints between batches was≥0.981.The linear range ofβ,β’-Dimethylacrylalkannin was 40.48-202.40μg.Conclusions The methods of TLC identification,fingerprint and content determination are simple,reliable and stable,which can be used for the quality control of Zilian oil.
