摘要
目的确定龙血树叶总黄酮的最佳提取工艺,并研究其体外抗氧化性。方法分别考察龙血树叶中总黄酮的索氏提取、热回流提取和超声提取方法,结果热回流提取为最佳方法。通过单因素试验和正交试验优化,分析提取条件(乙醇浓度、料液比、提取温度、提取时间)对龙血树叶总黄酮提取率的影响。采用自由基清除法和还原能力评价法分析黄酮提取物的体外抗氧化能力。结果确定的最佳提取工艺条件为:乙醇浓度80%、料液比1:100(g:mL)、提取温度70℃、提取时间120 min。在最佳提取条件下,龙血树叶总黄酮的提取率为1.79%。体外抗氧化结果表明:龙血树叶中总黄酮的提取物具有较好清除DPPH·和·OH的能力,同时也有一定的还原能力。结论所用方法稳定可靠,可用于龙血树叶总黄酮的提取和抗氧化性的测定,为龙血树的开发和应用提供依据。
OBJECTIVE To optimize extraction process of total flavonoids from leaves of Dracaena angustifolia and to study antioxidant activities.METHODS The flavonoids in the leaves of D.angustifolia were extracted by Soxhlet extraction,ultrasonic extraction and thermal reflux extraction,respectively.It was explored that thermal reflux extraction was the best.Through single factor and orthogonal optimization,the effects of extraction conditions(ethanol concentration,solid-liquid ratio,extraction temperature,extraction time)on the extraction rate of flavonoids from leaves of D.angustifolia were analyzed.Finally,the antioxidant activities of flavonoids extract were analyzed with the indexes of free radical scavenging ability and reducing ability.RESULTS The optimal extraction conditions were determined as follows:ethanol concentration of 80%,solid-liquid ratio of 1:100(g:mL),extraction temperature of 70℃,and extraction time of 120 minutes.Under the optimal extraction conditions,the extraction rate of flavonoids from leaves of D.angustifolia was 1.79%.The results of antioxidant experiments showed that the flavonoids from D.angustifolia had good ability to scavenge DPPH free radicals and hydroxyl radicals,and also had certain reducing ability.CONCLUSION This method is stable and reliable,and can be used for the extraction and antioxidant determination of flavonoids from leaves of D.angustifolia.It provides a theoretical basis for the further development and application of D.angustifolia.
