慢性鼻-鼻窦炎伴鼻息肉患者病变组织中IL-2和IL-35表达水平及其与Th1/Th2/Th17细胞因子的关系 被引量:18

摘要 目的分析慢性鼻-鼻窦炎伴鼻息肉(CRSwNP)患者病变组织中白介素-2(IL-2)、IL-35的表达水平及其与Th1/Th2/Th17细胞因子水平的关系。方法选取2017年6月—2019年6月收治的63例CRSwNP患者(CRSwNP组)及30例单纯鼻中隔偏曲患者(对照组)为研究对象。利用ELISA法检测CRSwNP组鼻息肉组织及对照组下鼻甲黏膜组织内IL-2、IL-35、γ干扰素(IFN-γ)、IL-4、IL-17的表达水平,并利用Pearson相关性分析鼻息肉组织中IL-2、IL-35水平与IFN-γ、IL-4、IL-17的相关性。结果CRSwNP组鼻息肉组织中IL-2、IL-35、IFN-γ水平低于对照组下鼻甲黏膜组织,IL-4、IL-17水平高于对照组下鼻甲黏膜组织(P<0.01);轻度CRSwNP患者鼻息肉组织中IL-2、IL-35、IFN-γ水平高于重度患者,IL-4、IL-17水平低于重度患者(P<0.01)。IL-2与IFN-γ呈正相关(r=0.482,P<0.01),与IL-4、IL-17呈负相关(r=-0.457、-0.542,P均<0.01)。IL-35与IFN-γ呈正相关(r=0.508,P<0.01),与IL-4、IL-17呈负相关(r=-0.471、-0.615,P均<0.01)。结论CRSwNP患者鼻息肉组织中IL-2、IL-35水平降低,可能与持续性Th2/Th17细胞激活,Treg细胞抑制有关。 Objective To analyze the expression levels of interleukin-2(IL-2)and IL-35 in lesion tissues and their relationship with levels of Th1/Th2/Th17 cytokines in patients with chronic rhinosinusitis with nasal polyps(CRSwNP).Methods A total of 63 patients with CRSwNP(the CRSwNP group)and 30 patients with simple nasal septum deviation(the control group)admitted from June 2017 to June 2019 were selected as the research subjects.The expression levels of IL-2,IL-35,interferon-γ(IFN-γ),IL-4 and IL-17 in nasal polyp tissues of CRSwNP group and inferior turbinate mucosa of control group were detected by ELISA,and the correlation of the levels of IL-2,IL-35 with IFN-γ,IL-4 and IL-17 in nasal polyp tissues was analyzed by Pearson correlation.Results The levels of IL-2,IL-35 and IFN-γin nasal polyps of CRSwNP group were lower than those in inferior turbinate mucosa of control group,and the levels of IL-4 and IL-17 were higher than those in inferior turbinate mucosa of control group(P<0.01).The levels of IL-2,IL-35 and IFN-γin nasal polyps of patients with mild CRSwNP were higher than those of patients with severe CRSwNP,and the levels of IL-4 and IL-17 were lower than those of patients with severe CRSwNP(P<0.01).IL-2 was positively correlated with IFN-γ(r=0.482,P<0.01)and negatively correlated with IL-4 and IL-17(r=-0.457,-0.542,both P<0.01).IL-35 was positively correlated with IFN-γ(r=0.508,P<0.01)and negatively correlated with IL-4 and IL-17(r=-0.471,-0.615,both P<0.01).Conclusion The levels of IL-2 and IL-35 in nasal polyps of CRSwNP patients were decreased,which might be related to persistent Th2/Th17 cell activation and Treg cell inhibition.
