摘要
目的分析人字草原植物中山奈酚和槲皮素化学成分。方法采用高效液相色谱法(HPLC)对人字草进行山奈酚和槲皮素含量的测定。色谱柱C18(4.6 mm×250 mm,5μm),流速1 ml/min,以甲醇-0.2%磷酸(65:35)为流动相,检测波长为360 nm。结果山奈酚回归方程:Y=10 496X-15.78,r=0.999 6,线性范围0.067~1.20 mg。槲皮素回归方程:Y=16625X-42.332,r=0.994,线性范围0.021~1.12mg。山柰酚的含量分别为0.679%、0.694%、0.698%、0.684%,平均含量为0.689%。槲皮素含量为0.315%、0.324%、0.321%、0.330%,平均含量为0.322%。HPLC结果表明人字草中山奈酚成分含量(0.068 mg/ml)1 ml成良好线性关系(r=0.999 6),平均回收率为100.04%,相对标准偏差(RSD)为1.1%(n=5);槲皮素成分含量(0.021 8 mg/ml)1 ml成良好线性关系(r=0.999 6),平均回收率为99.36%,RSD为0.97%(n=5)。结论此方法简便、重现性良好、结果准确可靠,可用于槲皮素和山柰酚含量测定。
Objective To analyze the chemical constituents of naphtenol and quercetin from kummerowia. Methods The content of kaempferol and quercetin was determined by high performance liquid chromatography(HPLC). Column C18(4.6 mm×250 mm,5 μm) with a flow rate of 1 ml/min. Methanol-0.2% phosphoric acid(65:35) was used as the mobile phase with a detect wavelength of 360 nm. Results Kaempferol regression equation: Y=10 496 x-15.78, r=0.9996, linear range 0.067-1.20 mg. quercetin regression equation: Y=16 625 X-42.332, r=0.994, linear range 0.021-1.12 mg. The content of kaempferol was 0.679%, 0.694%, 0.698%, 0.684%, and the average content was 0.689%. The contents of quercetin were 0.315%, 0.324%, 0.321%, 0.330%, and the average content was 0.32%. The results of HPLC showed that the content of Naempferol(0.068 mg · ml-1) 1 ml had a good linear relationship(r=0.9996), the average recovery was 100.04%, the RSD was 1.1%(n=5), the content of quercetin(0.0218 mg · ml-1) 1 ml had a good linear relationship(r=0.9996), the average recovery was 99.36%, and the RSD was 0.97%(n=5). Conclusion This method is simple, reproducible, accurate and reliable, and can be used for the determination of quercetin and kaempferol content.
