摘要
内生细菌是影响植物宿主生长和发育的重要因素,目前,苎麻中具有促生潜力的内生细菌鲜见报道。该研究从表面消毒的苎麻根部进行内生细菌的分离纯化,根据16S rRNA基因序列对所分离菌株进行系统发育分析,并从溶磷能力、固氮能力及产ACC脱氨酶潜力三个方面评估了内生细菌的促生长特性。结果表明,从苎麻根部共分离出12株细菌,经鉴定8株细菌属于Pseudomonas frederiksbergensis,其余4株细菌分属于Pseudomonas umsongensis、Pseudomonas sp.、Bacillus solani、Lysinibacillusxylanilyticus。促生特性试验表明,10株细菌具有溶磷能力,11株细菌具有固氮能力,但所分离内生细菌的产ACC脱氨酶潜力不高。苎麻根部内生细菌具有良好的溶磷、固氮等促生长特性,在农业微生物单一或复合菌肥开发利用方面具有潜在的应用价值。
Endophytic bacteria are an important factor affecting the growth and development of plant hosts. However, the endophytic bacteria with growth-promoting potential in ramie are rarely reported at present. In this study, endophytic bacteria were isolated and purified from surface sterilized roots of ramie, and the phylogenetic analysis of the strains was conducted by 16 S rRNA gene sequences, then the growth-promoting characteristics of the endophytic bacteria were evaluated from three aspects: the ability to dissolve phosphorus, the ability to fix nitrogen and the potential to produce ACC deaminase. Results showed that a total of 12 strains were isolated from ramie roots, and 8 strains were Pseudomonas frederiksbergensis, 1 strain was Pseudomonas umsongensis, 1 strain was Pseudomonas sp., 1 strain was Bacillus solani, 1 strain was Lysinibacillusxylanilyticus. Growth-promoting experiments showed that 10 strains had the phosphorus solubility, 11 strains had the function of nitrogen fixation. However, the ACC deaminase activities of the isolated endophytic bacteria were not high. In conclusion, endophytic bacteria isolated from ramie roots have significant growth-promoting characteristics, such as phosphate-dissolution and nitrogen-fixation, which have potential application value in exploitation and utilization of single or compound agricultural microbial fertilizer.
