摘要
目的研究JTE-013对博来霉素诱导的小鼠肺纤维化模型中肥大细胞及其细胞因子表达的变化。方法将40只健康清洁级雄性C57BL/6小鼠随机均分为8组:生理盐水对照组1周组、4周组(C1周组、C4周组);BLM模型组1周组、4周组(B1周组和B4周组);生理盐水+JTE-013对照组1周组、4周组(C+J 1周组和C+J 4周组);博来霉素+JTE-013组1周组、4周组(B+J1周组和B+J 4周组),每组5只,分别于第7天、28天处死。应用HE染色和Masson染色观察小鼠肺组织炎症及肺纤维化情况;通过甲苯胺蓝染色和迪夫快速染色观察肺组织及支气管肺泡灌洗液中(BALF)的肥大细胞的表达情况;ELISA方法检测BALF上清液中细胞因子IL-4、IL-5、TGF-β1及TNF-α的表达水平和血清中的IgE、组胺及S1P表达水平的变化。结果 JTE-013显著降低BLM诱导的小鼠肺组织细胞外胶原沉积;减少小鼠肺组织中肥大细胞浸润和BALF中肥大细胞的数量;减少BALF中IL-4、IL-5、TGF-β1及TNF-α的表达;降低血清中的IgE、组胺及S1P表达水平。结论 JTE-013可通过减少肥大细胞在肺内的浸润及其细胞因子的表达,减轻博来霉素诱导小鼠的肺纤维化。
To investigate the effects of JTE-013 on mast cells and their cytokine expression in mice with bleomycin(BLM)-induced pulmonary fibrosis, 40 specific pathogen-free healthy male C57 BL/6 mice were randomly divided into 8 groups: normal saline control group for 7 days, 28 days(1 week of group C, 4 weeks of group C), BLM model group for 7 days, 28 days(1 week of group B, 4 weeks of group B), normal saline + JTE-013 control group for7 days、28 days(1 week of group C+J, 4 weeks of group C+J), bleomycin + JTE-013 group for 7 days, 28 days(1 week of group B+J, 4 weeks of group B+J), with 5 mice in each group. Mice were sacrificed on the 7 th and 28 th day,respectively. HE staining and Masson staining were used to observe the inflammation and pulmonary fibrosis of lung tissue in mice;toluidine blue staining and Diff-Quik staining were used to observe the expression of mast cells in lung tissue and bronchoalveolar lavage fluid(BALF);ELISA was used to detect the expression levels of cytokines IL-4, IL-5, TGF-β1 and TNF-α in BALF supernatant and the change in the expression levels of IgE, histamine and S1 P in serum. Data showed that JTE-013 significantly decreased BLM-induced extracellular collagen deposition in mouse lung tissue, reduced mast cell infiltration in mouse lung tissue, and reduced the number of mast cells in BALF. JTE-013 also down-regulated the expression of IL-4, IL-5, TGF-β1 and TNF-α in BALF, and decreased IgE, histamine and S1 P expression levels in serum. Taken together, JTE-013 attenuates bleomycininduced pulmonary fibrosis in mice by reducing mast cell infiltration in the lung and down-regulating the expression of its cytokines.
