摘要
建立了高效液相色谱法测定注射用三磷酸胞苷二钠含量的方法。采用紫外-可见分光光度法(波长280 nm)测定总核苷酸含量,高效液相色谱法测定三磷酸胞苷二钠质量比。采用Agilent C_(18)柱,以十八烷基硅烷键合硅胶为填充剂,检测波长为270 nm。结果显示,建立的方法系统适用性、专属性、精密度和准确度均符合要求,在相对质量分数80%~120%内线性关系良好。该测定方法操作简便、准确,可用于三磷酸胞苷二钠的含量测定。
A method for the determination of cytidine disodium triphosphate for injection by high performance liquid chromatography(HPLC)was established.The total nucleotide content was determined by ultraviolet-visible spectrophotometry at 280 nm,and the mass ratio of cytidine disodium triphosphate was determined by HPLC.The Agilent C18 column was used with octadecylsilane chemically bonded silica as the filler,and the detection wavelength was 270 nm.The results showed that the system suitability,specificity,precision and accuracy of this method met the requirement,and it was linear in the relative mass fraction ranged from 80%to 120%.The method is simple and accurate,which can be used for the determination of cytidine disodium triphosphate.
