摘要
目的探讨保肾通络方含药血清对于高糖培养下足细胞p38通路及凋亡的作用。方法足细胞分为正常对照组、高糖组、保肾通络方含药血清低、中、高剂量组。CCK-8法检测各组足细胞活力,WB检测足细胞P-p38及p38蛋白表达,免疫荧光、RT-PCR检测足细胞凋亡相关蛋白caspase-3蛋白及mRNA表达,Hochest33258染色检测足细胞凋亡情况。结果与正常对照组相比,高糖组足细胞活力明显降低(P <0.05),与高糖组相比,保肾通络方含药血清组细胞活力明显升高(P <0.05)。与正常对照组相比,高糖组足细胞P-p38蛋白表达明显上调(P <0.05),与高糖组相比,保肾通络方含药血清组P-p38蛋白表达明显下调(P <0.05)。与正常对照组相比,高糖组足细胞caspase-3蛋白及mRNA表达明显上调(P <0.05),与高糖组相比,保肾通络方含药血清组足细胞caspase-3蛋白及mRNA表达明显下调(P <0.05)。与正常对照组比较,高糖组足细胞凋亡数目明显升高(P <0.05),与高糖组比较,保肾通络方含药血清组足细胞凋亡数目明显降低(P <0.05)。结论保肾通络方含药血清能够抑制高糖培养下足细胞p38通路激活,减轻足细胞凋亡。
Objective To observe the effect of drug-containing serum of Baoshen Tongluo granules(BSF) on podocyte apoptosis in high-glucose environment. Methods Differentiated mouse podocytes in vitro were divided into following groups: normal control group, high glucose group and drug-containing serum of BSF group. Cell viability was detected by CCK-8 assay. Cell apoptosis was detected by Hochest 33258 staining. Western blot were used for detecting protein expression of P-p38 and p38 in our study. Immunofluorescence and RT-PCR were used for detecting caspase-3 protein and mRNA expression. Results compared with NC group, cell viability of HG group was significantly decreased(P<0.05);P-p38 and caspase-3 expression was significantly increased(P<0.05);podocyte apoptosis was significantly increased(P<0.05). Compared with HG group, cell viability of BSF group was significantly increased(P<0.05);P-p38 and caspase-3 expression was significantly decreased(P<0.05);podocyte apoptosis was significantly decreased(P<0.05). Conclusion drug-containing serum of BSF significantly inhibited p38 pathway and podocyte apoptosis in high-glucose environment.
