温肾复甲汤对妊娠期甲状腺功能减退大鼠甲状腺功能及妊娠结局的影响

Effect of Wenshen Fujia decoction on thyroid function and pregnancy outcome in rats with hypothyroidism during pregnancy

摘要 目的观察温肾复甲汤对妊娠期甲状腺功能减退(以下简称甲减)大鼠甲状腺功能及妊娠结局的影响。方法将30只SD孕鼠按照随机数字表法分为空白组、模型组及温肾复甲汤低、中、高剂量组,每组6只。除空白组外,其余4组自妊娠第1 d(GD1)起予0.1%丙基硫氧嘧啶(PTU)溶液灌胃诱导并维持妊娠期甲减模型。GD11起,温肾复甲汤低、中、高剂量组分别予温肾复甲汤混悬液14.175、28.35、56.7 g/(kg·d)灌胃,空白组和模型组每日予2.1 mL/(0.2 kg·d)0.9%氯化钠注射液灌胃,各组均灌胃9d。GD20取血,化学发光法检测各组孕鼠血清促甲状腺激素(TSH)、游离甲状腺素(FT4)、游离三碘甲状腺原氨酸(FT3);取双侧子宫,计算胚胎吸收率。结果与空白组比较,模型组、温肾复甲汤低剂量组孕鼠血清TSH升高(P<0.05),FT4、FT3降低(P<0.05),温肾复甲汤中剂量组FT3降低(P<0.05);与模型组比较,温肾复甲汤低、中、高剂量组TSH均降低(P<0.05),FT4均升高(P<0.05),温肾复甲汤中、高剂量组FT3升高(P<0.05);与温肾复甲汤低剂量组比较,温肾复甲汤中、高剂量组TSH均降低(P<0.05),FT4均升高(P<0.05),温肾复甲汤高剂量组FT3升高(P<0.05);与温肾复甲汤中剂量组比较,温肾复甲汤高剂量组FT3升高(P<0.05)。与空白组比较,模型组及温肾复甲汤低剂量组孕鼠胚胎吸收率均升高(P<0.05);与模型组比较,温肾复甲汤中、高剂量组孕鼠胚胎吸收率降低(P<0.05);与温肾复甲汤低剂量组比较,温肾复甲汤中、高剂量组孕鼠胚胎吸收率均降低(P<0.05)。结论温肾复甲汤可改善妊娠期甲减大鼠的甲状腺功能及妊娠结局,且有一定的量效关系。 Objective To observe the effect of Wenshen Fujia decoction on thyroid function and pregnancy outcome in rats with hypothyroidism during pregnancy.Methods 30 pregnant SD rats were randomly divided into blank group,model group and Wenshen Fujia decoction low,middle and high dose groups,with 6 rats in each group.Except the blank group,the other four groups were given 0.1%propylthiouracil(PTU)solution by gavage from the first day of gestation(GD1)to induce and maintain the hypothyroidism model of pregnancy.From GD11 on,Wenshen Fujia decoction low,meiddle and high dose groups were given Wenshen Fujia decoction suspension 14.175,28.35 and 56.7 g/(kg·d)by gavage respectively,while the blank group and model group were given 2.1 mL/(0.2 kg·d)0.9%sodium chloride injection daily.All groups were given gavage for 9 days.The serum thyroid stimulating hormone(TSH),free thyroxine 4(FT 4)and free triiodothyronine(FT 3)were detected by chemiluminescence method at GD20,and bilateral uterus was taken to calculate embryo absorption rate.Results Compared with the blank group,the serum TSH of pregnant rats in the model group and the low dose Wenshen fujia decoction group was increased(P<0.05),while FT 4 and FT 3 were decreased(P<0.05),and the FT 3 of the middle dose Wenshen Fujia decoction group was decreased(P<0.05).Compared with the model group,TSH was decreased in the low,middle and high dose groups of Wenshen Fujia decoction(P<0.05),FT 4 was increased(P<0.05),and FT 3 was increased in the middke and high dose groups of Wenshen Fujia decoction(P<0.05).Compared with the low dose Wenshen Fujia decoction group,TSH was decreased in the middle and high dose groups of Wenshen Fujia decoction(P<0.05),and FT 4 was increased(P<0.05),and FT 3 in the high dose Wenshen Fujia decoction group was increased(P<0.05).Compared with the middle dose group of Wenshen Fujia decoction,the FT 3 of the Wenshen Fujia decoction high dose group was increased(P<0.05).Compared with the blank group,the embryo absorption rate of pregnant mice in the model group and the low dose Wenshen Fujia decoction group was increased(P<0.05).Compared with the model group,the embryonic absorption rate of pregnant mice in the middle and high dose Wenshen Fujia decoction group was decrease(P<0.05).Compared with the low dose group of Wenshen Fujia decoction,the embryo absorption rate of pregnant rats in the middle and high dose Wenshen Fujia decoction groups was decreased(P<0.05).Conclusion wenshenfujia decoction can improve the thyroid function and pregnancy outcome of hypothyroidism rats,and there is a certain dose effect relationship.
