柔肝补肾酒对腺嘌呤致肾阳虚大鼠睾酮、雌二醇水平及睾丸、精囊腺病理变化的影响 被引量:2

Effects of Nourishing Liver&Tonifying Kidney Wine(柔肝补肾酒)on Testosterone,Estradiol Levels and Pathological Changes of Testis and Seminal Vesicle in Rats with Kidney-Yang Deficiency Induced by Adenin

摘要 目的:观察柔肝补肾酒对腺嘌呤诱导肾阳虚大鼠模型血清睾酮、雌二醇、肾功能及睾丸、精囊腺病理变化的影响。方法:0.7%腺嘌呤饲料制备肾阳虚模型,将30只雄性Wistar大鼠随机分为正常组,模型组,白酒对照组,柔肝补肾酒高、低剂量组,每组6只,给药组每日给予相应剂量药物灌胃,隔天1次,共14 d。观察大鼠体质量的变化,应用ELISA法测定大鼠血清睾酮(T)、雌二醇(E 2);应用全自动生化分析仪测定血清尿素氮(BUN)、天门冬氨酸氨基转移酶(AST);光镜下观察大鼠睾丸、精囊腺组织病理变化。结果:与正常组比较,模型组大鼠体质量显著减轻,血清T、E 2水平显著降低,BUN水平显著升高;与模型组比较,柔肝补肾酒(高、低)剂量组大鼠体质量显著升高,T、E 2水平显著升高,BUN水平显著降低(P<0.01,P<0.05)。各组大鼠血清AST水平无明显差异。病理观察结果显示:模型组大鼠睾丸生精小管生殖细胞数量减少,细胞排列紊乱,小管腔内间质出现空隙,腔内成熟精子数量显著减少,间质细胞数量减少,小管变形部分塌陷;精囊腺上皮细胞增生、空泡变性。柔肝补肾酒高、低剂量组病理改变明显轻于模型组。结论:柔肝补肾酒对腺嘌呤致肾阳虚大鼠睾丸和精囊腺的损伤有保护作用,其机制可能与上调T、E 2水平,下调BUN水平有关。 Objective:To investigate the effects of Nourishing Liver&Tonifying Kidney Wine(NLTKW)on testosterone,estradiol,BUN and pathological changes of testis and seminal vesicle in rats with kidney-yang deficiency induced by adenine.Methods:Mixed feedstuff contained 7%adenine was used to establish model.30 Wistar rats were randomly divided into normal group,model group,white liquor control group and NLTKW high-dose and low-dose groups,with 6 rats in each group.Medication groups were given relevant medicine for gavage,very other day,14 days.Rats’body weight changes were recorded,the levels of blood testosterone(T)and estradiol(E 2)were measured by ELISA,the level of blood urea nitrogen(BUN)and aspartate aminotransferase(AST)were measured by automatic biochemistry analyzer.The histopathology of the testis and seminal vesicle was observed by light microscope.Results:Compared with normal group,the weight of rats in the model group was reduced,the levels of blood T and E 2 were lower,the level of blood BUN was higher;Compared with model group,the weight of rats in the NLTKW high-dose and low-dose group was increased,the levels of blood T and E 2 were higher,the level of blood BUN was lower(P<0.01,P<0.05);there was no significant difference in blood AST levels among groups.Pathological observation showed that in the model group,the number of germ cells in testicular seminiferous tubules was decreased,the arrangement of cells was disordered,the tubule lumen occurred interstitial space,the number of mature sperm in the lumen decreased significantly,the number of interstitial cells decreased,and the tubule deformed part collapsed,the epithelial cell of seminal vesicle proliferated.Pathological changes in the NLTKW high-dose and low-dose group were significantly lighter than those in the model group.Conclusion:NLTKW can reduce the injury of testis and seminal vesicle in rats with kidney-yang deficiency induced by adenine and has protective effects on the rat model through increasing the levels of T and E 2 and reducing the level of BUN.
