摘要 目的研究以卡介苗为载体的细粒棘球绦虫egG1Y162融合蛋白(rBCG-egG1Y162)免疫小鼠的免疫应答改变,评价其在小鼠实验中发挥的动物免疫保护作用,为研究新型有针对性的囊型包虫病疫苗奠定基础。方法免疫保护性评价实验分为3组:rBCG-egG1Y162组、卡介苗(BCG)组和正常对照(NS)组,用重组疫苗免疫Balb/c小鼠,肌肉注射疫苗第8周后用攻虫实验感染各组一半数量小鼠,在第24周后获取小鼠相应的器官和血清标本。酶联免疫吸附试验(enzyme linked immunosorbent assay,ELISA)检测血清中IgG亚类IgG1、IgG2a、IgG2b和IgE抗体水平变化,评价疫苗的体液免疫水平。计算小鼠感染模型中细粒棘球蚴的包囊数量及其大小,对包囊湿重进行称量,评价该重组疫苗对小鼠的免疫保护性。结果与BCG组和NS组相比,rBCG-egG1Y162免疫组小鼠血清中IgG2a、IgG2b和IgE抗体水平增高,差异有统计学意义(P<0.05),IgG1抗体水平降低,差异有统计学意义(P<0.05)。各组小鼠在免疫后被动感染棘球蚴,rBCG-egG1Y162免疫组具有78.1%的囊泡抑制率,Eg囊湿重与BCG组和NS组相比差异有统计学意义(P<0.05),结论以BCG为载体的rBCG-egG1Y162疫苗免疫小鼠后,攻虫实验中的囊泡抑制率显著增高,小鼠体内寄生虫的数量也显著减少,初步认为rBCG-egG1Y162可以作为囊型包虫病的候选疫苗进行深入研究。 Objective To study the immune response of mice immunized with the vaccine of rBCG-egG1Y162 of Echinococcus granulosus(Eg),and evaluate its immunological protection of the vaccine on mice,and laid a foundation for constructing a new and efficient vaccine against Cystic echinococcosis(CE).Methods The mice used in the immune protection evaluation test were divided into three groups:rBCG-egG1Y162 group,BCG group and normal control group(NS).Balb/c mice were intramuscularly injected with vaccine.Half of the mice in each group were infected with protoscolex at 8 weeks after immunization.The serum levels of IgG,subclasses IgG1,IgG2a,IgG2b and IgE were detected by Enzyme linked immunosorbent assay(ELISA).The number,size and wet weight of Echinococcus cysts in the mouse infection model were recorded.Results Compared with BCG group and NS group,the serum level of IgG2a,IgG2b and IgE in rBCG-egG1Y162 immune group was increased significantly(P<0.05),and the IgG1 antibody level was decreased significantly(P<0.05).The inhibition rate of cysts in the rBCG-egG1Y162 group was 78.1%,and the wet cyst weight in the rBCG-egG1Y162 group was less than the one in the BCG group and NS group with statistically significant(P<0.05).Conclusion The rBCG-egG1Y162 vaccine could effectly inhibit the infection of Echinococcus in the mice,and might be a candidate for development of Echinococcosis vaccine.
