摘要
目的观察补肾化痰法对肥胖型多囊卵巢综合征(PCOS)雌鼠子宫内膜VEGF/Akt通路相关分子表达的影响,探讨该法对子宫内膜的影响。方法采用7~8周龄SD雌鼠,来曲唑加高脂乳剂建立肥胖型PCOS模型,以补肾化痰法经典方治疗,结合阴道脱落细胞涂片、血清性激素和卵巢形态学进行模型验证,ELISA法检测子宫匀浆VEGF含量,免疫组织化学染色法检测子宫组织E2R、VEGFR2,Western blotting检测子宫组织p-PDK1、p-Akt蛋白表达水平。结果与空白对照组相比,模型对照组大鼠孕激素显著下降(P<0.01),VEGF、VEGFR2、p-PDK1显著升高(P<0.01),E2R升高(P<0.05),p-Akt表达上调,提示造模成功,子宫内膜可能与VEGF/Akt通路相关;与模型对照组相比,中药高剂量组大鼠P升高(P<0.05),E2R显著下降(P<0.01),VEGF、VEGFR2、p-PDK1下降(P<0.05),p-Akt表达降低。结论VEGF/Akt通路可调控肥胖型PCOS雌鼠的子宫内膜,补肾化痰法能有效下调相关分子的表达,抑制VEGF/Akt通路进而改善肥胖型PCOS子宫内膜微环境。
Objective To observe the effect of kidney-nourishing and phlegm-removing method(KNPR)on the expression of VEGF/Akt pathway in the endometrium of obese PCOS female rats and explore the effect of this method on the regulation of endometrium.Methods We chose 7~8 week sold female SD rats and treated them with the method of KNPR after modeling by Letrozole,high-fat and cholesterol.The models were verified by the smears of vaginal exfoliated cells,the serum sex hormone levels and the morphological changes of ovarian.VEGF concentration in uterine homogenatewas detected by ELISA method and the immunohistochemical method waschosen to detect the expressions of E2R and VEGFR2 in the uterus tissue.The expressions of phosphorylated PDK1 and phosphorylated Akt were detected by Western blotting.Results Compared with the blank control group,the P level of the model group had decreased significantly(P<0.01).The expressions of VEGF,VEGFR2 and PDK1 in the uterus tissue of the model group were increased significantly(P<0.01).E2R wasincreased(P<0.05)and the expressions of Akt was higher.It suggested that the success of the model and the decrease of edometrium might be related to the VEGF/Akt pathway.Compared with the model group,the P level of the traditional Chinese medicine high dose groupwasincreased(P<0.05).The expression of E2R wasdecreased significantly(P<0.01),the expressions of VEGF,VEGFR2 and PDK1 were decreased(P<0.05)and the expressions of Akt was lower.Conclusion The VEGF/Akt pathway regulates the endometrium of obese PCOS female rats.It is speculated that the method of KNPR could effectively down-regulate the expressions of relevant molecules in the endometrium and further affect the proliferation and differentiation of the molecular microenvironment of the endometrium by inhibiting the VEGF/Akt pathway.
