摘要 目的:探讨血管内皮生长因子(VEGF)在根尖周囊肿骨破坏过程中的作用。方法:免疫组织化学染色检测32例根尖周囊肿囊壁中VEGF表达及微血管密度,分析其与骨破坏的关系;分离囊壁及正常间充质对照成纤维细胞,ELISA和RT-qPCR检测其中VEGF表达。条件培养基诱导RAW264.7细胞破骨向分化,贝伐单抗阻断VEGF,计数TRAP阳性破骨细胞。结果:囊壁中VEGF呈强阳性,其表达及微血管密度与骨破坏面积正相关。囊壁成纤维细胞VEGF及破骨诱导效率显著高于对照,并受贝伐单抗抑制(P<0.05)。结论:根尖周囊肿囊壁VEGF能促血管增生及破骨细胞分化,导致骨破坏。 Objective: To investigate the effect of vascular endothelial growth factor(VEGF) on bone destruction of radicular cysts. Methods: The expression of VEGF and microvessels density in 32 cases of radicular cysts were investigated with immunohistochemical staining. Fibroblasts were isolated from the capsule of radicular cyst and the normal mesenchymal controls. The expression of VEGF in the 2 groups were detected by ELSA and RT-qPCR. Conditioned medium was made of supernatants of the cells and was used to induce the cell line RAW264.7, and bevacizumab was added to block VEGF. TRAP positive osteoclasts were counted. Results: VEGF was strongly expressed in the capsule of radicular cyst. Its expression and microvessel density were positively correlated with the bone destruction area. The fibroblasts from radicular cysts exhibited significantly higher expression of VEGF and induced more osteoclasts, in contrast with the controls. However, the formation of osteoclasts could be significantly inhibited by bevacizumab(P<0.05). Conclusion: High expression of VEGF in the capsule of radicular cyst can lead to bone destruction by promoting vascular proliferation and inducing osteoclast differentiation.
