摘要
通过碱性艳蓝B与美司那的作用,建立了测定药物及人血样品中美司那的高灵敏瑞利光散射(RLS)新方法。在pH=9.25的B-R碱性溶液中,美司那与碱性艳蓝B以静电引力相作用生成一种蓝色的二元离子缔合物,产生以355 nm为特征峰的瑞利光散射增强信号,其体系的瑞利光散射增强强度(ΔI RLS)与美司那的质量浓度在0.005~0.16 mg/L范围呈线性关系,检出限为0.0046 mg/L。该方法简便、快速、灵敏度高,用于市售药物及人体血液中美司那的测定,回收率和相对标准偏差(%,n=6)分别为98.3%~102%和1.8%~2.5%。
A Rayleigh light scattering method was established for the determination of mesna in drugs and human blood samples through the reaction of basic brilliant blue B and mesna.In an alkaline solution of pH 9.25 B-R buffer,mesna reacted with basic brilliant blue B to form a blue binary ionic associate complex by electrostatic attraction,which generated Rayleigh light scattering(RLS) enhanced signal with a characteristic peak of 355 nm.The Rayleigh light scattering enhancement intensity(ΔIRLS) of the system was linear with mesna’s mass concentration(0.005-0.16 mg/L),and the detection limit was 0.0046 mg/L.The method was simple,rapid and highly sensitive.It has been applied to the determination of mesna in commercially available mesna drugs and human blood with the recovery rates and relative standard deviations(n=6) of 98.3% to 102% and 1.8% to 2.5%,respectively.
