PB试验结合BBD响应面法优化冬虫夏草菌菌丝体多糖的提取工艺 被引量:6

Optimization of Polysaccharide Extraction of Ophiocordyceps sinensis Mycelium by Plackett-Burman Design Combined with Box-Behnken Response Surface Methodology

摘要 以青海省冬虫夏草菌(Ophiocordyceps sinensis)菌丝体为材料,多糖得率为指标,优化冬虫夏草菌菌丝体多糖提取工艺。在单因素试验基础上,利用Plackett-Burman试验筛选得到显著影响因素,再运用响应面法对提取工艺参数进行优化,应用相同试验设计对热水浸提和微波提取工艺进行比较。结果表明热水浸提下冬虫夏草菌菌丝体多糖提取率低于微波提取,最佳微波提取工艺参数为:目数为80目,料液比为1∶26 g·mL^-1,浸提次数为3次,微波功率为330 W,微波时间为4 min,冬虫夏草菌菌丝体多糖得率达9.06%。 With Ophiocordyceps sinensis mycelium as raw material and polysaccharide yield as index of evaluation,to compare and optimize the processing of polysaccharide extraction.Principal factors related to extraction rate of polysaccharide were optimized by the Plackett-Burman design,based on this study through response surface analysis the optimum extraction conditions.Use the same experimental design to compare microwave extraction and hot water extraction.The results showed that the microwave extraction rate of polysaccharides was higher.The optimal microwave extraction conditions were mesh number was 80,solid-liquid ratio was 1∶26,extraction frequency was 3 times,microwave power was 330 W,microwave time was 4 min and the polysaccharide yield of O.sinensis mycelium reached 9.06%.
