中华白僵菌产胞内粗多糖提取及发酵条件优化 被引量:1

Optimization of extraction and fermentation of intracellular crude polysaccharides from Beauveria

摘要 本文采用单因素试验、正交实验、Plackett-Burman实验、最陡爬坡实验以及Box-Behnken实验对中华白僵菌(Beauveria sinensis)产孢内粗多糖浸提条件与发酵条件进行优化。结果显示,经正交实验优化后,中华白僵菌产胞内粗多糖提取含量提升37.0%,经Box-Behnken实验优化后确定最佳发酵条件为:可溶性淀粉添加量5%、酵母浸膏添加量2.5%、装液量100 mL/250 mL、初始pH值5.5、温度27℃、转速160 r/min,多糖最高产量为(31.5±1.0)mg/g,对比优化前提高了57.5%。 The extraction and fermentation conditions of crude polysaccharides from Beauveria sinensis were optimized by single factor and orthogonal experiments,Plackett-Burman experiment,steepest climbing test and Box-Behnken experiment.The results showed that the yield of crude polysaccharide extracted from Beauveria sinensis increased by 37.0% with the optimization by orthogonal experiment.The optimum fermentation conditions by Box Benhnken experiment were:soluble starch 5%,yeast extract 2.5%,the liquid volume 100 mL/250 mL,initial pH 5.5,temperature 27℃,and the rotation speed 160 r/min.The highest polysaccharide yield was(31.5±1.0)mg/g,which was 57.5% higher than that before the optimization.
