摘要
对珍稀食药用菌虎奶菇(Pleurotus tuber-regium)菌丝体培养条件进行优化,并通过MTT试验验证了其菌丝体提取物对肺癌细胞增殖的影响。结果发现,其液体培养基中大豆饼粉的浓度为10 g·L^-1、pH为6.8,培养时间为168 h时,液体培养获得的菌丝体干重为17.5 g·L^-1;虎奶菇乙酸乙酯及正丁醇提取物浓度在6.25μg·mL^-1和800.00μg·m L^-1时,对肺癌细胞增殖的抑制率最高分别为84.51%和93.13%,计算IC50值分别为252.62μg·m L^-1和108.25μg·mL^-1。
Pleurotus tuber-regium is a rare edible and medicinal fungus.The mycelium culture conditions were optimized,and the effect of mycelium extracts on lung cancer cell proliferation was studied by MTT experiments.The results found that when the concentration of soybean meal in the liquid medium was 10 g·L^-1,the pH was 6.8,and the cultivation time was 7 days,the dry weight of the mycelium obtained was 17.5 g·L^-1.The concentration of ethyl acetate extract is in the range of 6.25μg·mL^-1-800μg·mL^-1,and the inhibition rate on lung cancer cell proliferation are up to 84.51%and 93.13%respectively.The calculated IC50 value was 252.62μg·mL^-1 and 108.25μg·mL^-1.
